Gua Sha, de traditionele Chinese therapeutische huid schrapen, veroorzaakt onderhuidse microvasculaire bloed extravasatie. Wij rapporteren een protocol van bioluminescentie beeldvorming van HO-1-<em> Luciferase</em> Transgene muizen aan te tonen dat Gua Sha heem upregulates oxygenase-1 (HO-1) in meerdere organen.
Gua Sha is een traditionele Chinese folk therapie die de huid heeft schrapen om onderhuidse microvasculaire bloed extravasatie en blauwe plekken veroorzaken. Het protocol voor bioluminescente optische beeldvorming van HO-1-<em> Luciferase</em> Transgene muizen die in dit handschrift geeft een snelle<em> In vivo</em> Test van de opregulatie van het heem oxygenase-1 (HO-1) genexpressie in reactie op de Gua Sha procedure. HO-1 is al lang bekend dat cytoprotection te bieden tegen oxidatieve stress. De opregulatie van HO-1, beoordeeld door de bioluminescentie output, wordt gedacht aan een anti-oxidatieve reactie op circulerende hemoglobine producten vrijgegeven door Gua Sha vertegenwoordigen. Gua Sha werd toegediend door herhaalde slagen van een gladde lepel voorsprong op gesmeerd huid op de rug of andere gerichte lichaamsdeel van de transgene muis tot petechiën (splinterbloedingen) of bloeduitstortingen (blauwe plekken) indicatie van de extravasatie van bloed van subcutane haarvaten waargenomen. Na Gua Sha, werden bioluminescentie beeldvorming sessies dagelijks uitgevoerd gedurende enkele dagen aan de dynamiek van HO-1 expressie te volgen in meerdere interne organen.
Transcriptie van HO-1, een induceerbare vorm van heem oxygenase, wordt opgereguleerd door vele factoren, waaronder heem, waterstof peroxide, UV-straling, hypoxie, en lichamelijke spanningen. Het gehele lichaam, zoals de gerapporteerde HO-1 bioluminescentie-protocol zorgt voor een snelle momentopname van systemische HO-1 expressie in meerdere organen.
