Optische resolutie fotoakoestische microscopie (OR-PAM) is een opkomende technologie die in staat van beeldvorming optische absorptie contrasten<em> In vivo</em> Met cellulaire resolutie en gevoeligheid. Optische microscopie, waardevolle inzichten op cellulair en organellen niveaus, is alom erkend als een mogelijk biomedische technologie. Als de pijlers van de in vivo drie-dimensionale (3-D) optische microscopie, single-/multi-photon fluorescentie microscopie en optische coherentie tomografie (OCT) hebben aangetoond hun buitengewone gevoeligheid voor fluorescentie-en optische verstrooiing contrasten, respectievelijk. Echter, de optische absorptie contrast van biologische weefsels, die essentiële fysiologische / pathologische informatie codeert, nog niet toetsbaar.
De opkomst van biomedische photoacoustics heeft geleid tot een nieuwe tak van optische microscopie optische resolutie fotoakoestische microscopie (OR-PAM) 1, waar de optische bestraling is gericht op de diffractielimiet om cellulaire 1 of zelfs 2 subcellulaire niveau laterale resolutie te bereiken. Als een waardevolle aanvulling op de bestaande optische microscopie-technologieën, OR-PAM brengt in ten minste twee nieuwigheden. Eerst en vooral, OR-PAM detecteert optische absorptie in contrast met buitengewone gevoeligheid (dat wil zeggen 100%). De combinatie van OR-PAM met fluorescentie microscopie 3 of met optische-verstrooiing op basis van 04 oktober (of met beide) biedt uitgebreide optische eigenschappen van biologische weefsels. Ten tweede, OR-PAM codeert optische absorptie in akoestische golven, in tegenstelling tot de pure optische processen in fluorescentie microscopie en oktober, en biedt achtergrond-free detectie. De akoestische detectie in OR-PAM mitigeert de effecten van optische verstrooiing van signaal degradatie en natuurlijk elimineert mogelijke storingen (dat wil zeggen, CROSSTALKS) tussen excitatie en detectie, dat is een veel voorkomend probleem bij fluorescentie microscopie te wijten aan de overlap tussen de excitatie-en fluorescentie spectra.
Uniek voor optische absorptie beeldvorming, heeft OR-PAM aangetoond breed biomedische toepassingen sinds de uitvinding, met inbegrip van maar niet beperkt tot, neurologie 5, 6, oogheelkunde 7, 8, vasculaire biologie 9, 10 en dermatologie. In deze video, leren wij de systeemconfiguratie en de uitlijning van de OR-PAM evenals de experimentele procedures voor in-vivo functionele microvasculaire beeldvorming.
In deze video geven we een gedetailleerde instructie over de experimentele protocollen van OR-PAM, met inbegrip van de systeemconfiguratie, systeem uitlijning en typische experimentele procedures. Label-free, niet-invasieve OR-PAM heeft ingeschakeld studies van microvasculaire functioneren en de stofwisseling op een enkele capillair basis en daardoor heeft het potentieel om uit te breiden ons begrip van de microcirculatie-gerelateerde fysiologie en pathologie.
