I denna rapport beskriver vi hur ytplasmonresonans används för att upptäcka toxin inträde i den mottagande cytosolen. Denna mycket känsliga metoden kan ge kvantitativa uppgifter om mängden cytosolic toxin, och den kan tillämpas på en rad olika gifter.
AB gifter består av en enzymatisk En subenhet och en cell-bindande B-subenhet 1. Dessa gifter utsöndras i den extracellulära miljön, men de agerar på mål inom eukaryota cytosolen. Vissa AB gifter resa med vesikler transportörer från cellytan till endoplasmatiska retiklet (ER) innan cytosolen 2-4. I ER, för att den katalytiska En kedja dissocierar från resten av toxinet och rör sig genom en protein-ledande kanalen nå sitt cytosoliskt mål 5. Den flyttas, cytosoliskt En kedja är svåra att upptäcka eftersom toxin människohandel till ER är en mycket ineffektiv process: de flesta internaliseras toxin är dirigeras till lysosomer för nedbrytning, så bara en liten del av ytbundet toxin når Golgiapparaten och ER 6 -12.
För att övervaka toxin translokation från ER till cytosolen i odlade celler, kombinerade vi en subcellulära fraktionering protokoll med highly känslig metod för ytplasmonresonans (SPR) 13-15. Membranet på toxin-behandlade celler är selektivt permeabilized med digitonin, vilket gör att insamling av en cytosoliskt bråkdel som sedan perfusion över en SPR-sensor belagda med ett anti-toxin En kedja antikropp. De antikroppsförsedda sensorn kan fånga och upptäcka pg / ml mängder cytosoliska toxin. Med detta protokoll är det möjligt att följa kinetiken av toxin inträde i cytosolen och för att karakterisera hämmande inverkan på translokation evenemanget. Koncentrationen av cytosolic toxin kan också beräknas från en standardkurva som genereras med kända mängder av en kedja standarder som har perfusion över sensorn. Vår metod är en snabb, känslig och kvantitativ detektionssystem som inte kräver radioaktiv inmärkning eller andra ändringar till målet toxin.
