Vi presenterar en metod för att bilda en avbildning fönster i mus skalle som sträcker sig över millimeter och är stabil under månader utan inflammation av hjärnan. Denna metod lämpar sig väl för longitudinella studier av blodflödet, cellulära dynamik, och cell / vaskulär struktur genom att använda två-foton-mikroskopi.
Avbildning in vivo av kortikalt funktion kräver optisk tillgång till hjärnan utan störning av den intrakraniala miljön. Vi presenterar en metod för att bilda en polerad och förstärkt förtunnad skallen (portar) fönster i musen skallen som spänner över flera millimeter i diameter och är stabil i månader. Skallen förtunnas till 10 till 15 pm i tjocklek med en handburen borrmaskin för att uppnå optisk klarhet, och därefter täcktes med cyanoakrylatlim och ett täckglas för att: 1) ger styvhet, 2) inhibera benåterväxt och 3) reducera ljusspridning av oegentligheter på benet ytan. Eftersom skallen inte överskrids, är någon inflammation som kan påverka processen som studeras kraftigt. Bildgivande djup på upp till 250 pm under den kortikala ytan kan åstadkommas med användning av två-foton-mikroskopi laserskanning. Detta fönster är väl lämpad för att studera cerebralt blodflöde och cellulär funktion i både sövda och vakna beredningar. Det erbjuder ytterligare ophet att manipulera cellaktivitet med användning optogenetics eller att störa blodflödet i målinriktade kärl genom bestrålning av cirkulerande fotosensibilisatorer.
Två-foton-avbildning genom en PORTS fönster kräver transmission genom den förtunnade benet och dura, som dämpar det laserljus och adderar optiska aberrationer på större djup 8.
