Specifika bakterier finns i association med djurvävnad 1-5. Även om mycket uppmärksamhet har ägnats åt patogena interaktioner är lite känt om de processer som styr den reproducerbara förvärvet av nyttiga / bakteriefloran från omgivningen. Den Ijus-organet mutualism mellan den marina gramnegativ bakterie V. fischeri och Hawaiian bobtail squid, E. scolopes representerar en mycket specifik växelverkan där en värd (E. scolopes) fastställs ett symbiotiskt förhållande med endast en bakterieart (V. fischeri) under hela dess livstid 6,7. Bioluminiscens produceras av V. fischeri under denna interaktion ger en anti-aggressiv nytta för E. scolopes under nattliga aktiviteter 8,9, medanden näringsrika värdvävnad ger V. fischeri med en skyddad nisch 10. Under varje värd generation är detta förhållande rekapituleras, vilket motsvarar en förutsägbar process som kan bedömas i detalj i olika stadier av symbiotisk utveckling. I laboratoriet, om uppsamlades under de första 30-60 minuter och överfördes till symbiont-fritt vatten till juvenil bläckfisk luckan aposymbiotically (uncolonized), och, inte kan koloniseras förutom genom den experimentella inokulum 6. Här beskriver vi en metod för att bedöma graden av kolonisering som uppstår när nykläckta aposymbiotic E. scolopes utsätts för (konstgjord) havsvatten innehållande V. fischeri. Denna enkla analys beskriver ympning, naturlig infektion och återvinningav den bakteriella symbiont från den framväxande ljuset organ E. scolopes. Man ser till att ge en enhetlig miljö för djuren under symbiotisk utveckling, särskilt med avseende på vattenkvalitet och signaler ljus. Metoder för att karakterisera det symbiotiska befolkningen som beskrivs inkluderar (1) mätning av bakteriellt härrörande mareld, och (2) direkt koloniräkning av återvunnet symbionter.
Koloniseringen analysen som beskrivs möjliggör analys av en naturlig symbiotiskt process i en kontrollerad laboratoriemiljö. Som sådan kan den användas för att bedöma kolonisering av mutanta stammar, från olika naturliga isolat, och under olika kemiska system.
