Det generelle princip ved denne fremgangsmåde er at måle membran deformation som reaktion på veldefineret undertryk påføres gennem en mikropipette til celleoverfladen. Denne fremgangsmåde tilvejebringer et effektivt værktøj til at studere biomekaniske egenskaber af substrat-bundne celler.
Stigende antal undersøgelser viser, at biomekaniske egenskaberne af de enkelte celler spiller vigtige roller i flere cellulære funktioner, herunder celleproliferation, differentiering, migration og celle-celle-interaktioner. De to nøgleparametre for cellulære biomekanik er cellulære deformerbarhed eller stivhed og cellernes evne til at optage og generere kraft. Her beskriver vi en hurtig og enkel metode til at anslå celle stivhed ved måling af graden af membranens deformation som reaktion på negativt tryk fra en glasmikropipette til celleoverfladen, en teknik, der kaldes Mikropipette Aspiration eller Microaspiration.
Microaspiration udføres ved at trække en glaskapillær for at skabe en mikropipette med en meget lille spids (2-50 um diameter afhængigt af størrelsen af en celle eller en vævsprøve), som derefter forbundet med en pneumatisk tryktransducer og bragt til afslutning nærhed af en celle under et mikroskop. Nårspidsen af pipetten rører en celle, et trin med negativt tryk tilføres til pipetten ved det pneumatiske tryk transducer genererer veldefineret tryk på cellemembranen.
