In this protocol, we describe methods to efficiently transfect murine macrophage cell lines with siRNAs using the Amaxa Nucleofector 96-well Shuttle System, stimulate the macrophages with lipopolysaccharide, and monitor the effects on inflammatory cytokine production.
Macrophages are key phagocytic innate immune cells. When macrophages encounter a pathogen, they produce antimicrobial proteins and compounds to kill the pathogen, produce various cytokines and chemokines to recruit and stimulate other immune cells, and present antigens to stimulate the adaptive immune response. Thus, being able to efficiently manipulate macrophages with techniques such as RNA-interference (RNAi) is critical to our ability to investigate this important innate immune cell. However, macrophages can be technically challenging to transfect and can exhibit inefficient RNAi-induced gene knockdown. In this protocol, we describe methods to efficiently transfect two mouse macrophage cell lines (RAW264.7 and J774A.1) with siRNA using the Amaxa Nucleofector 96-well Shuttle System and describe procedures to maximize the effect of siRNA on gene knockdown. Moreover, the described methods are adapted to work in 96-well format, allowing for medium and high-throughput studies. To demonstrate the utility of this approach, we describe experiments that utilize RNAi to inhibit genes that regulate lipopolysaccharide (LPS)-induced cytokine production.
In dit protocol beschrijven we efficiënte methoden om genen in muizen macrofaag cellijnen met siRNA remmen en de effecten van de behandelingen die de aangeboren immuunrespons. Deze procedures worden uitgevoerd in 96-well formaat, waardoor RNAi schermen in middel- of high-throughput wijze.
In reactie op infectie, mens zet onmiddellijk aangeboren immuunrespons en een trager, maar specifieker adaptieve immuunrespons. Deze snelle aangeboren immuunrespons impliceert de werving en activering van fagocyterende aangeboren immuuncellen waaronder macrofagen 1. Klassiek geactiveerde macrofagen betrokken zijn bij acute ontstekingsreacties en produceren antimicrobiële eiwitten en verbindingen, cytokinen en chemokinen, en presenteren antigenen 2,3. Alternatief geactiveerde macrofagen een rol spelen bij het reguleren van de immuniteit, het handhaven tolerantie, weefselherstel en wondgenezing 4-8. Vanwege hun gemak functies, Macrofagen een rol in talrijke ziekten zoals atherosclerose, artritis, kanker en 9 spelen. Ondanks hun belang in de aangeboren immuunrespons, kan macrofagen uitdagend cellen om mee te werken. In het bijzonder is het moeilijk om doelmatige transfectie met lipide reagentia in macrofagen te verkrijgen zonder geassocieerde toxiciteit 10,11. Zelfs wanneer siRNA efficiënt afgeleverd macrofagen, de robuustheid van RNAi-geïnduceerde gen vaak knockdown kan tamelijk gematigd zijn en kunnen variëren van gen tot gen.
Om deze technische uitdagingen te overwinnen, hebben wij transfectie en knock-down technieken 12-16 geoptimaliseerd in twee muis macrofaag cellijnen, RAW264.7 17 en J774A.1 18. Deze aanpak maakt gebruik van de Amaxa Nucleofector 96-well Shuttle Systeem voor transfectie; dit systeemeen combinatie van speciale reagentia en elektroporatie cellen in 96-well formaat 19 transfecteren. Na transfectie, beschrijven we methoden om cel herstel en de levensvatbaarheid te maximaliseren en de daaropvolgende siRNA-geïnduceerde gen knockdown maximaliseren. Ter illustratie van de bruikbaarheid van deze benadering beschrijven we een protocol voor siRNA levering aan deze macrofaag cellijnen, stimuleren deze cellen met lipopolysaccharide (LPS), en bewaken de aangeboren immuunrespons op het productieniveau van verscheidene pro-inflammatoire cytokines.
