We describe a protocol for modifying cell affinity of a scaffold surface in aqueous environment. The method takes advantage of titanium dioxide photocatalysis to decompose organic film in the photo-irradiated region. We show that it can be used to create microdomains of scaffolding proteins, both ex situ and in situ.
Organische verontreinigingen geadsorbeerd op het oppervlak van titaniumdioxide (TiO2) kan worden afgebroken door fotokatalyse onder ultraviolet (UV) licht. Hier beschrijven we een nieuw protocol toepassen van de TiO 2 fotokatalyse lokaal veranderen cell affiniteit van het substraatoppervlak. Voor dit experiment werd een dunne TiO 2-film werd sputter coating op een glazen dekglaasje en de TiO 2 oppervlak werd vervolgens gemodificeerd met een organosilaan monolaag afgeleid van octadecyltrichloorsilaan (OTS), die celadhesie remt. Het monster werd ondergedompeld in een celkweekmedium en gerichte UV licht werd bestraald met een achthoekige gebied. Wanneer een neuronale cellijn PC12-cellen werden uitgeplaat op het monster cellen gehecht alleen de UV-bestraalde gebied. We tonen verder aan dat deze modificatie van het oppervlak kan ook worden uitgevoerd in situ, dat wil zeggen, zelfs wanneer de cellen groeien op het substraat. Een juiste modificatie van het oppervlak vereist een extracellulaire matrix pProtein collageen aanwezig in het medium ten tijde van UV bestraling. De hier gepresenteerde techniek kan eventueel worden toegepast in patroonvorming meerdere celtypen te construeren gemengde cultuursystemen of willekeurig manipuleren cellen onder kweek.
Halfgeleiderlithografie processen en zijn derivaten – zoals fotolithografie 1,2, electron-beam lithografie 3-6 en microcontact afdrukken 7-10 – zijn inmiddels uitgegroeid tot een gevestigde praktijk in de celbiologie aan levende cellen te groeien in een bepaalde positie en geometrie. De patroon werkwijze berust op het gebruik van microfabricated substraten bestaande uit micro-eiland cel permissief coating in een niet-permissieve achtergrond. Dergelijke substraat dient als een template patroon van de cellen. Deze technologieën hebben ons de nieuwe werkwijzen om cellen en hun functie ingenieur bij enkel- en meercellige level, uittreksel de intrinsieke eigenschappen van de cellen, en de doorvoer van celgebaseerde drugonderzoek 11 verhogen.
De mate-van-vrijheid in cel patroonvorming zou enorm toenemen als het patroon geometrie in situ kan worden veranderd, dat wil zeggen, terwijl cellen gekweekt op de sppervlakte. De conventionele werkwijzen voor patroonvorming niet direct hier toegepast, aangezien samples verwerken atmosfeer of in vacuüm. Daarom diverse nieuwe oppervlakte modificatie technieken zijn voorgesteld, die gebaseerd zijn, bijvoorbeeld op fotoreactieve verbindingen 12,13 of laser ablatie 5,14, om er maar een paar te noemen. De voorgestelde werkwijzen zijn goed beoordeeld door Robertus et al. 15, en recenter door Choi et al. 16 en 17 Nakanishi.
Hier in dit artikel beschrijven we een nieuw protocol van in-situ modificatie van het oppervlak, die profiteren van de fotokatalytische afbraak van organische moleculen neemt een titaniumdioxide (TiO2) oppervlak 18,19. In deze werkwijze wordt een TiO 2 film ingevoegd tussen het glassubstraat en de organische film die de cellen interfaces en de organische film wordt ontleed in situ door het plaatselijk bestralen ultraviolette (UV)licht om een gebied van belang (λ <388 nm). We tonen aan dat het nieuwe protocol kan worden gebruikt om micropatterns van extracellulaire matrixeiwitten en levende cellen zowel ex situ als in situ te creëren. TiO 2 biocompatibel, chemisch stabiel, en optisch transparant, kenmerken waarvan maakt kinderen te voeren in cel-kweek-experimenten. Dit protocol voorziet in een materiaal-wetenschap gebaseerde alternatief voor het wijzigen van celcultuur steigers in celcultuur omgeving.
In onze huidige protocol, werd TiO 2 film gevormd door RF-magnetron sputteren. Wij zijn voorstander van deze methode van afzetting, omdat het ons toelaat om reproduceerbaar te bereiden een fotokatalytische TiO 2 film met een sub-nm ruwheid. Hoewel sputterdepositie processen zijn bekend om materialen wetenschappers en elektronische ingenieurs, kan het niet zijn vrij toegankelijk voor biologen.
