Dit protocol beschrijft de bereiding en karakterisering van een dendrimere magnetische resonantie imaging (MRI) contrastmiddel dat cyclen gebaseerde macrocyclische chelaten coördinerende paramagnetische gadolinium-ionen draagt. In een serie MRI-experimenten in vitro, deze agent produceerde een versterkte MRI signaal in vergelijking met de commercieel beschikbare monomere analoog.
Paramagnetische complexen van gadolinium (III) met acyclische of macrocyclische chelaten zijn de meest gebruikte contrastmiddelen (CA) voor magnetische resonantie beeldvorming (MRI). Hun doel is om de versoepeling snelheid van water protonen in het weefsel te versterken, waardoor de MR beeldcontrast en de specificiteit van de MRI-metingen toeneemt. Huidige klinisch goedgekeurde contrastmiddelen vanaf hoogmoleculaire moleculen die snel uit het lichaam geklaard. Het gebruik van dendrimeren als dragers van paramagnetische chelaten kunnen een belangrijke rol spelen in de toekomstige ontwikkeling van efficiëntere MRI contrastmiddelen. Met name de toename van lokale concentratie van het paramagnetische soorten resulteert in een hoger signaal contrast. Bovendien is dit CA levert een langere retentietijd tissue tijd vanwege het hoge molecuulgewicht en grootte. Hier tonen we een geschikte procedure voor de bereiding van macromoleculaire MRI contrastmiddelen op basis van poly (amidoamine) (PAMAM) dendrimeren monomacrocyclische DOTA-type chelatoren (DOTA – 1,4,7,10-tetraazacyclododecaan-1,4,7,10-tetraacetaat). De chelerende eenheid werd toegevoegd door het benutten van de reactiviteit van het isothiocyanaat (NCS) groepen naar het oppervlak amine- groepen van de PAMAM dendrimeer te thioureum brugvorming. Dendrimere producten werden gezuiverd en geanalyseerd door middel van kernmagnetische resonantie spectroscopie, massaspectrometrie en elementanalyse. Tenslotte werden hoge resolutie MR beelden die zijn opgenomen en het signaal contrasten verkregen uit de voorbereide dendrimere en in de handel verkrijgbare monomeer agenten werden vergeleken.
Magnetic resonance imaging (MRI) is een krachtige en niet-ioniserende beeldvormingstechniek schaal gebruikt in biomedisch onderzoek en klinische diagnostiek vanwege de invasieve aard en uitstekende intrinsieke zacht weefsel contrast. De meest gebruikte MRI methoden gebruiken het signaal verkregen uit water protonen leveren van hoge resolutie beelden en informatie binnen de weefsels op basis van verschillen in de dichtheid van het water signalen. De signaalintensiteit en de specificiteit van de MRI-experimenten kan verder worden verbeterd middels contrastmiddelen (CA). Dit zijn paramagnetische of superparamagnetische soorten die invloed hebben op de longitudinale (T1) en transversale (T2) relaxatie maal respectievelijk 1,2.
Complexen van het lanthanide-ion gadolinium met polyaminozuren polycarbonzuur liganden de meest gebruikte T 1 CA's. Gadolinium (III) verkort de T 1 ontspanningtijd van water protonen, waardoor het signaal contrast in MRI-experimenten 3 toeneemt. Echter, ionische gadolinium giftig; de grootte benadert die van calcium (II), en ernstige gevolgen calcium bijgestaan signalering in cellen. Daarom worden acyclische en macrocyclische chelaten gebruikt om deze toxiciteit te neutraliseren. Verschillende multidentaat liganden zijn tot nu toe ontwikkeld, waardoor gadolinium (III) complexen met een hoge thermodynamische stabiliteit en kinetische inertheid 1. Op basis van het 12-ledige azamacrocycle cyclen, met name het tetracarbonzuur afgeleide DOTA (1,4,7,10-tetraazacyclododecaan-1,4,7,10-acetaat) zijn de meest onderzochte en toegepaste complexen van deze CA klasse.
Toch GdDOTA-type CA's zijn lage gewicht systemen moleculair, het weergeven van bepaalde nadelen zoals een lage contrast efficiency en een snelle renale excretie. Macromoleculaire en multivalente CA kan een goede oplossing voor deze problemen 4 zijn. Sinds CA biodistributie wordt vooral bepaald door de grootte, macromoleculaire CA tonen veel langere retentietijden in de weefsels. Even belangrijk is de meerwaardigheid van deze middelen resulteert in een verhoogde lokale concentratie van de monomere MR probe (bijvoorbeeld GdDOTA complex), fundamentele verbetering van de verworven MR-signaal en het meten kwaliteit.
Dendrimeren behoren tot de meest geprefereerde draagstructuren voor de bereiding van meerwaardige CA voor MRI 4,5. Deze sterk vertakte macromoleculen met goed gedefinieerde afmetingen zijn gevoelig voor verschillende koppelingsreacties te binden. In dit werk, melden wij de voorbereiding, zuivering en karakterisering van een dendrimere CA voor MRI, bestaande uit een generatie 4 (G4) poly (amidoamine) (PAMAM) dendrimeer gekoppeld aan GdDOTA-achtige chelaten (DCA). We beschrijven de synthese van de reactieve DOTA-derivaat en de koppeling met de PAMAM dendrimeer. Bij complexering met Gd (III), de standaard fysisch-chemische karakterisering procedure van DCA werd uitgevoerd. Tenslotte werden MRI experimenten uitgevoerd om het vermogen van DCA MR beelden te produceren met een sterker contrast dan die verkregen uit laagmoleculaire CA tonen.
Voorbereiding van de dendrimere MRI-contrastmiddel vereist juiste keuze van de monomere eenheid (dat wil zeggen, de chelator voor Gd (III)). Ze verminderen de toxiciteit van dit paramagnetische ion en tot nu toe een groot aantal acyclische en macrocyclische chelatoren dienen daartoe 1-3.
