We describe here a method for regulating picornavirus tropism by incorporating sequences complementary to specific microRNAs into the viral genome. This protocol can be adapted to all different classes of viruses with modifications based upon the length and nature of their life cycle.
Cell-specific restriction of viral replication without concomitant attenuation can benefit vaccine development, gene therapy, oncolytic virotherapy, and understanding the biological properties of viruses. There are several mechanisms for regulating viral tropism, however they tend to be virus class specific and many result in virus attenuation. Additionally, many viruses, including picornaviruses, exhibit size constraints that do not allow for incorporation of large amounts of foreign genetic material required for some targeting methods. MicroRNAs are short, non-coding RNAs that regulate gene expression in eukaryotic cells by binding complementary target sequences in messenger RNAs, preventing their translation or accelerating their degradation. Different cells exhibit distinct microRNA signatures and many microRNAs serve as biomarkers. These differential expression patterns can be exploited for restricting gene expression in cells that express specific microRNAs while maintaining expression in cells that do not. In regards to regulating viral tropism, sequences complementary to specific microRNAs are incorporated into the viral genome, generally in the 3′ non-coding regions, targeting them for destruction in the presence of the cognate microRNAs thus preventing viral gene expression and/or replication. MicroRNA-targeting is a technique that theoretically can be applied to all viral vectors without altering the potency of the virus in the absence of the corresponding microRNAs. Here we describe experimental methods associated with generating a microRNA-targeted picornavirus and evaluating the efficacy and specificity of that targeting in vitro. This protocol is designed for a rapidly replicating virus with a lytic replication cycle, however, modification of the time points analyzed and the specific virus titration readouts used will aid in the adaptation of this protocol to many different viruses.
Utvecklingen av en mångsidigt användbar, enkel och effektiv metod för att konstruera en vektor med begränsad tropism ger en stor möjlighet att öka säkerheten, biologisk förståelse och terapeutisk användbarhet av virus. Flera mekanismer för att rikta viral tropism inklusive transductional, transkriptions och translationella baserade tekniker. Men dessa metoder är i allmänhet inte är tillämpliga på alla vektorsystem, kan kräva defekta signalvägar i målceller eller kräver införande av stora kodande sekvenser i det virala genomet. Dessutom kan dessa metoder resulterar i attenuering av viruset, vilket avsevärt hindrar deras terapeutiska aktivitet och begränsa insikt i den omodifierade systemet.
MicroRNAs är små (22-25 nukleotider), icke-kodande RNA som förmedlar geners uttryck i eukaryota celler. MicroRNAs funktion genom att binda komplementära målsekvenser (responselement) i budbärar-RNA (mRNA) resulti ng i avskrift destabilisering, nedbrytning eller translationella repression. MicroRNAs binder normalt svarselement med partiell komplementaritet och ger små ändringar i genuttryck 1, 2, 3, 4, 5. Mer betydande förändringar i genuttryck kan åstadkommas genom att öka kompletterar svarselementet 6. Tusentals mogna mikroRNA har identifierats i ett antal olika arter och många uppvisar differentiella uttrycksmönster i en mängd olika cell- och vävnadstyper 7, 8, 9. Dessa mikroRNA signaturer kan utnyttjas för cellspecifik begränsning av virusamplifiering genom att införliva perfekt komplementära responselement i det virala genomet 10,= "xref"> 11, 12, 13. Det övergripande målet för denna microRNA inriktning teknik är att styra tropism av en vektorgenomet utan ytterligare dämpning.
Användbarheten av denna metod för att reglera viral tropism ursprungligen visats i lentivirala vektorer för att begränsa transgent uttryck i specifika vävnader 14, 15, 16. Denna teknik har sedan använts på ett brett spektrum av replikerande och icke-replikerande virala vektorer för förbättrad genterapi samt att förbättra säkerhetsprofilen för många onkolytiska virus genom att eliminera oönskade toxicitet i normala vävnader 10, 11, 12, 13, 17 . Det har också använts för att skapa säker och effective levande försvagade vacciner samt att förbättra virus och vaccintillverkningsprocesser 18, 19, 20, 21. MicroRNA inriktning av en vektor kan möjliggöra dämpning hos vaccinerade värdar eller riktade system med bibehållen vildtyp tillväxtnivåer i producentsystem. MicroRNA inriktning kan också användas för att förbättra biosäkerhet av virus för forskningsändamål genom att begränsa överföring i en art (t.ex. människa) med bibehållen överföring i andra värdar 22. Slutligen microRNA inriktning kan möjliggöra fördjupade analyser av virus livscykler och specifika roller celltyper i patogenes och immunitet genom segregerande virustillväxt 23, 24, 25, 26.
Denna teknik erbjuder en alternative inriktningsmetod som lätt genomförs och tillämpas för alla virussystem. De största begränsningarna av denna teknik inbegriper trial and error optimering, potentialen för utrymnings mutationer, och potentiella ej åsyftade effekter på endogena transkript. Emellertid kan dessa begränsningar i allmänhet övervinnas med optimerat och rationell responselement design. Positiv-sense RNA-virus tenderar att vara särskilt mottaglig för microRNA-inriktning på grund av den positiva ense-riktning av deras genom och tillgången av transkripten till mikroRNA maskiner under helt cytoplasmatisk replikationscykeln.
