Dette manuskript beskriver det nye setup og betjening af en photoacoustic mikroskopi og optisk kohærens tomografi dual-modalitet system for noninvasive, etiket-fri chorioretinal billeddannelse af større dyr som kaniner.
Photoacoustic okulær imaging er en spirende oftalmologiske imaging-teknologi, der kan noninvasively visualisere øjenvæv ved at konvertere lysenergi til lydbølger og er i øjeblikket under intensiv efterforskning. Men de fleste rapporterede hidtidige arbejde er fokuseret på billeddannelse af bageste segment af øjnene af små dyr, som rotter og mus, som udgør en udfordring for kliniske menneskelig oversættelse på grund af små øjeæblet størrelser. Dette manuskript beskriver en roman photoacoustic mikroskopi (PAM) og optisk kohærens tomografi (OCT) dual-modalitet system for bageste segment billeddannelse af øjnene af større dyr som kaniner. Systemkonfiguration, system justering, animalsk forberedelse og dual-modalitet eksperimentelle protokoller for in vivo, noninvasive, etiket-fri chorioretinal imaging i kaniner er detaljerede. Effektiviteten af metoden, der er påvist gennem repræsentative eksperimentelle resultater, herunder retinal og choroidal Vaskulaturen fremstillet ved PAM og OCT. Håndskriftet indeholder en praktisk vejledning til at gengive de imaging resultater i kaniner og fremme photoacoustic okulær imaging i større dyr.
De seneste årtier har været vidne til den eksplosive udvikling af inden for biomedicinsk photoacoustic billeddannelse1,2,3,4,5,6,7 ,8. Baseret på energi konverteringen af lys til lyd, den nye photoacoustic imaging kan visualisere biologiske prøver på skalaer fra organeller, celler, væv, organer til små dyr hele kroppen og kan afsløre sin anatomiske, funktionelle, molekylære, genetiske, og metaboliske oplysninger1,2,9,10,11,12. Photoacoustic imaging har fundet unikke applikationer i en række biomedicinsk områder, såsom celle biologi13,14, vaskulære biologi15,16, neurologi17,18 , onkologi19,20,21,22, dermatologi23, farmakologi24og hæmatologi25,26. Dens anvendelse i oftalmologi, dvs photoacoustic okulær imaging, har tiltrukket betydelig interesse fra både forskere og klinikere og er i øjeblikket under aktiv efterforskning.
I modsætning til anvendes rutinemæssigt okulær tænkelig teknologier27, såsom fluorescein angiografi (FA) og indocyanine grønne angiografi (ICGA) (baseret på fluorescens kontrast), optisk kohærens tomografi (OCT) (baseret på optiske spredning kontrast) , og dens afledte OCT angiografi (baseret på bevægelse kontrast af røde blodlegemer), photoacoustic okulær imaging bruger optisk absorption som kontrast mekanisme. Dette adskiller sig fra konventionelle okulær billeddannelsesteknologier og giver et unikt værktøj til at studere optisk absorption egenskaber i øjet, hvilket som regel er forbundet med den patofysiologiske status af øjenvæv28. Til dato har betydelige er fremragende arbejde blevet gjort i photoacoustic okulær imaging29,30,31,32,33,34,35, 36,37, men disse undersøgelser fokuserer på det bageste segment af øjnene af små dyr, som rotter og mus. De banebrydende studier godt demonstrere mulighederne for photoacoustic billeddannelse i oftalmologi, men der er stadig lang vej at gå mod kliniske oversættelse af teknologi siden øjeæblet størrelser af rotter og mus er meget mindre (mindre end en tredjedel) end af mennesker. På grund af udbredelsen af ultralyd bølger over et væsentligt længere afstande lider signalet intensitet og billedkvalitet høj grad når teknikken bruges til billeddannelse den bageste segment af større øjne.
Mod dette mål, vi for nylig rapporteret noninvasive, etiket-fri chorioretinal imaging i levende kaniner ved hjælp af integreret photoacoustic mikroskopi (PAM) og spektral-domæne OCT (SD-OCT)38. Systemet har fremragende ydeevne og kunne visualisere nethinden og årehinden i øjnene af større dyr baseret på endogene absorption og spredning kontrast af øjenvæv. Foreløbige resultater i kaniner viser at PAM noninvasively kunne skelne enkelte retinal og choroidal blodkar ved hjælp af en laser eksponering dosis (~ 80 nJ) betydeligt under den amerikanske nationale standarder Institute (ANSI) sikkerhed grænse (160 nJ) på 570 nm39; og OLT kunne klart løse forskellige retinal lag, årehinden og sclera. Det er den første demonstration af bageste segment billeddannelse af større dyr ved hjælp af PAM og kan være et stort skridt mod kliniske oversættelse af den teknologi, der overvejer at øjeæblet størrelsen af kaniner (18,1 mm)40 er næsten 80% af aksial længde mennesker (23,9 mm).
I dette arbejde, vi leverer en detaljeret beskrivelse af dual-modalitet billedbehandlingssystem og eksperimenterende protokoller bruges til noninvasive, etiket-fri chorioretinal imaging i levende kaniner og demonstrere ordning arbejdsindsats gennem repræsentative retinal og choroidal imaging resultater.
En intakt og regelmæssig tårefilmen er afgørende for høj kvalitet fundus billeder. En uregelmæssig og forringet tåre film kan betydeligt nedbrydes billed kvalitet42. For at bevare integriteten af tårefilmen og forhindre hornhinde overfladiske punktformet keratopati, er det kritisk at smøre hornhinden ved hjælp af øjenskyller meget ofte, ca hver to min.
