Abstract
日には、発達段階別にいくつかのタイプの幹細胞は、変性疾患の治療のための焦点。まだ、初期大規模な細胞死と治療効果は低いなどの特定の側面は、広範な臨床翻訳障害。移植前に幹細胞の遺伝子工学は、治療の幹細胞の効果を最適化する有望な方法として浮上しました。しかし、安全で効率的な遺伝子送達システムはまだ欠けています。したがって、適切な手法の開発には、幹細胞ベースの治療の現在の課題を解決するアプローチがあります。

Introduction
人間の歯小嚢は、発展途上の歯1,2を取り巻く緩やかな ectomesenchymally から派生した結合組織です。破骨細胞形成と骨延長法歯の噴火プロセスの調整するため、その機能の横には、この組織は、特に歯周組織3,4、5の開発のための茎および祖先のセルを隠し持っています。したがって、歯小嚢は成体幹細胞6,7を収穫するための代替ソースとしてと見なされます。

Protocol
HDFSCs は、ロストック大学医療センター歯科口腔外科学講座によって提供される抽出の知恵の歯の歯科小胞から分離されています。インフォームド コンセントと書面による承認は、すべての患者から得られました。この研究は、(許可番号 2017年-0158) ロストック大学の倫理委員会で承認されました。
1. hDFSCs の単離
注:親知らずは、細菌汚染を防ぐためには、抽出前にない噴出する必要があります。

必要な溶液の調製
- リン酸緩衝生理食塩水 (PBS) の準備/ペニシリン-ストレプトマイシン-グルタミン (P S G) ソリューション: 495 mL の PBS の P S G 溶液 5 mL を混ぜて。4 ° C で 50 mL の因数を格納します。
- HDFSC 培養液の準備: 445 mL 基本培地の 5 ml の抗菌剤、ウシ胎児血清 (FBS) の 50 mL を混ぜます。4 ° C でのストア
- コラゲナーゼ型を準備私貯蔵液 (30 mg/mL): 基礎培地 10 mL では 1% 抗菌剤を添加したコラゲナーゼ型の 300 mg を希釈します。積極的にソリューションをミックスします。0.2 μ m フィルターを使用して、ソリューションをフィルター処理します。コラゲナーゼを 500 μ l 添加のストア因数-20 ° C で保存溶液を型します。

手術の手順
- 十分な量を注入 (最大: 2 mL) 局所麻酔薬の。
- 作成し、三つどもえの骨膜を発生させます。舌のフラップを上げる必要はありません。

- 優しくラウンド ハード金属バリで頬側から遠位骨を加工します。

- 口腔内から歯卵胞を外し、PBS/P ・ S ・ G 溶液 50 mL の因数にそれらを置きます。

歯卵胞の酵素の消化力
- あらかじめ暖かい hDFSC 培と部屋の温度 (RT) PBS/P ・ S ・ G ソリューション。

- 25 cm2種細胞懸濁液は、細胞培養用フラスコと 37 ° C、5% CO2と 20% O2セルを孵化させなさい。
注:組織が完全に消化されていない場合は、細胞培養用フラスコ同様に残りの組織を転送します。 - 変更中慎重に 24 h 細胞播種後。その後媒体の confluency まで 3 日を変更します。
注:HDFSCs 細胞播種後プラスチック付着 24 h をする必要があり、媒体を変更するだけで非付着性のセルおよび血液成分から分離することができます。

セルを収穫
- あらかじめ暖かい hDFSC 培、PBS/P ・ S ・ G ソリューションと右にトリプシン/エチレンジアミン edta 溶液
- 培養用フラスコから上澄みを廃棄し、PBS/P ・ S ・ G 溶液 5 mL に合流するセルを洗浄します。
- 培養用フラスコに/トリプシン EDTA 溶液の 1 mL を追加し、37 ° C で 3 分間インキュベートHDFSC 培養液 3 mL を培養用フラスコに追加することによって消化力プロセスを停止します。
- 15 mL コニカル遠心チューブに細胞懸濁液を移すと破棄した上清で 353 x gで 10 分間の懸濁液を遠心分離し、再 hDFSC 培地の適切な量の餌を中断します。
- セルをカウント: の細胞懸濁液の 10 μ L を混ぜて軽くトリパン ブルー溶液 10 μ L。カウント室に 10 μ L を適用し、hDFSC 細胞量を計算します。
2. hDFSCs のキャラクタリゼーション

- 細胞のフローサイトメトリーによる解析のための準備
- 必要な溶液の調製
- PBS/EDTA (2 mM) の準備: 996 mL の PBS の EDTA (0.5 M) の 4 つの mL を混ぜて。
- 染色バッファーを準備: PBS/EDTA (2 mM) の 995 mL を混ぜて BSA の 5 g。0.22 μ m のフィルター ユニットを使用して、ソリューションをフィルターし、使用まで 4 ° C で保存します。
- パラホルムアルデヒド (PFA) 原液 (4%) を準備: 4 g PFA 100 mL の PBS を希釈し、80 ° C への解決策を熱ソリューションをミックスし、7.3 に pH 値を調整します。因数は、使用までそれぞれ金額 (1.5 mL)、-20 ° C で店で PFA ソリューションを得られます。
注意: PFA 煙は有毒なので、換気の発煙のフード PFA ソリューションを準備します。
- セル (1.4) を収穫後 1.5 mL 遠心チューブに 5 x 10 の4セルの 14 サンプルを転送します。4 ° C で 10 分間 300 x gで懸濁液を遠心分離します。上清を捨てます。

HDFSCs の多能性の微分の潜在性

3. hDFSCs のトランスフェクション

注:RNases からの汚染を防ぐためには、直接 RNase 除染液を用いたトランスフェクションの複合体の準備する前にワークスペースをクリーンします。RNase フリーの材料とソリューションのみを使用します。
注:むしろ影付きルームで次の作業を実行します。- ミール ストック溶液 (50 μ M) を調製: 再 Cy3 標識前駆体ミール (5 nmol) 100 μ L ヌクレアーゼ フリー水を中断します。因数は、使用までミールそれぞれ量 (5 μ L) の原液と-20 ° C で暗闇の中でストアを取得しました。

- 37 ° C、5% CO2、および 20% O2 24 時間で細胞を孵化させなさい。
4. 遺伝子の解析

細胞トランスフェクション後採取
注:同じ 15 mL 円錐形遠心分離機管の 1 つのサンプルのすべての携帯ソリューションを収集します。- トランスフェクション後、24 h は、それぞれ遠心チューブ内のサンプルの上澄みを収集します。
- 1 ml の PBS のセルを洗浄、それぞれ遠心管に PBS を転送、500 μ L を追加/トリプシン EDTA (RT) のセルにします。37 ° C で 3 分間ソリューションを孵化させなさい
- 培 (RT) の 1 つの mL を追加してセルとそれぞれ遠心管に転送ソリューションに trypsinization を停止します。4 ° C で 10 分間 300 × gで遠心分離細胞

細胞のフローサイトメトリーによる解析のための準備
- 上清を捨てます。再細胞染色バッファー (4 ° C) と 1.5 mL 遠心チューブに転送細胞液 100 μ L を中断します。

Representative Results
人間の歯小嚢の組織から hDFSCs を収穫する詳細な分離命令を紹介します。ルーチン手術中に歯小嚢への簡単アクセス、ため成体幹細胞の抽出のための有望なソースです。
孤立した hDFSCs は、MSCs13の定義に記述されているすべての特性を示した。実際には、細胞がプラスチック付着下培養条件を説明し、線維芽細胞のような形状 (図 1) が表示されます。フローサイトメトリーによる解析では、CD45 と CD117 は欠席 (図 2) (cd29)、CD44、CD73、CD90 CD105 を含むある特定の表面抗原のパネルを hDFSCs に表現を明らかにしました。また、脂肪細胞、骨および特定の in vitro における細胞の軟骨分化培養条件 (FABP4) 脂肪酸結合蛋白質 4 の染色、オステオカルシン、アグリカン (図 3) が確認されました。
説明カチオン性脂質ベースのトランスフェクション戦略には、ミール吸収性の生菌 24 h 後トランスフェクション (図 4 b) の ~ 100% の hDFSCs の効率的な一時的な遺伝的変更が有効になります。また、transfected (図 4 a) と融合 (図 4) サンプルは、穏やかな細胞処理条件を証明する死んだ細胞の対等な量を示した。

Discussion

MiRs を挿入することによって幹細胞の遺伝子工学は、低細胞生存43,55,56,57などの幹細胞を用いた治療の困難を克服するために新たな戦略です。このプロトコルは、市販のカチオン性脂質ベースのトランスフェクション試薬を用いた hDFSCs 合成ミールの効率的な導入の重要な手順を発表しました。多数の臨床試験58,59治療試薬、薬や核酸などの配信のためカチオン性リポソーム製剤の応用について調べた。カチオン性リポソームが、用量依存的で潜在的に細胞毒性遺伝子式59,60,61に細胞膜や変更の整合性への例えば,損傷を引き起こすことによって.したがって、特に注意はトランスフェクションによって細胞への毒性を支払われる必要があります。
特に、指定されたトランスフェクション条件は、効率性と細胞毒性に関して hDFSCs のミールを介した遺伝子組み換えの最適化されています。それにもかかわらず、他の研究を示した別のセル型62,63,に追加核酸、プラスミド DNA を含む mRNA や siRNA、配信するためこのトランスフェクション試薬の成功のアプリケーション64,65,66,67,68. これらの研究の結果は、理想的な配信条件が大幅に変化し、セル タイプごとに定義する必要があることを明らかにしました。
