Vi beskriver en protokoll for etikett-fri identifikasjon av lymfocytt subtyper kvantitative fase bildebehandling og en maskinlæring algoritme. Målinger av 3D brytningsindeks tomograms av lymfocytter presenterer 3D morfologiske og biokjemiske informasjon for enkeltceller som analyseres deretter med en apparat-innlæring algoritme for identifikasjon av celletyper.
Her beskriver vi en protokoll for etikett-fri identifikasjon av lymfocytt subtyper kvantitative fase bildebehandling og maskinlæring. Identifikasjon av lymfocytt subtyper er viktig for studiet av immunologi samt diagnose og behandling av ulike sykdommer. Foreløpig avhengige standard metoder for klassifisering lymfocytt merking bestemt membran proteiner via antigen-antistoff reaksjoner. Men bærer disse merking teknikker den potensielle risikoen ved å endre cellulære funksjoner. Protokollen beskrevet her overvinner disse utfordringene ved å utnytte innebygd optisk kontraster målt ved 3D kvantitative fase bildebehandling og en maskinlæring algoritme. Måling av 3D brytningsindeks (RI) tomograms av lymfocytter gir kvantitativ informasjon om 3D morfologi og fenotyper individuelle celler. De Biofysiske parameterne Hentet fra den målte 3D RI tomograms analyseres deretter kvantitativt med en maskin læring algoritme, aktivere etikett-fri identifikasjon av lymfocytt på en enkeltcelle nivå. Vi måle i 3D RI tomograms av B, CD4 + T og CD8 + T-lymfocytter og identifisert deres celletyper med over 80% nøyaktighet. I denne protokollen beskriver vi detaljerte trinn for lymfocytt isolasjon, 3D kvantitative fase bildebehandling og maskinlæring identifisere lymfocytt typer.
Lymfocytter kan klassifiseres i ulike undergrupper inkludert B, hjelper (CD4 +) T, cytotoksiske (CD8 +) T og regulatoriske T celler. Hver lymfocytt har en annen rolle i det adaptive immunsystemet; for eksempel, produserer B-lymfocytter antistoffer, mens T-lymfocytter gjenkjenner bestemte antigener, fjerne unormale celler og regulere B-lymfocytter. Lymfocytt funksjon og regulering er tett kontrollert av og relatert til ulike sykdommer, inkludert kreft1, autoimmune sykdommer2og virusinfeksjoner3. Identifikasjon av lymfocytter er derfor viktig å forstå sine patofysiologiske roller i slike sykdommer og for immunterapi i klinikker.
Foreløpig avhengig metoder for klassifisering lymfocytt antigen-antistoff reaksjoner av målretting bestemte overflaten membran proteiner eller overflate markører4. Målretting overflate markører er en presis og nøyaktig metode for å bestemme lymfocytt typer. Men krever det dyre reagenser og tidkrevende prosedyrer. Videre bærer risikoen for endring av membran proteinstrukturer og endring av cellulære funksjoner.
For å overvinne disse utfordringene, introduserer protokollen beskrevet her etikett-fri identifikasjon av lymfocytt 3D kvantitative fase imaging (QPI) og maskinen lære5. Denne metoden gjør det mulig for klassifisering av lymfocytt typer på en enkeltcelle nivå basert på morfologiske informasjon fra etikett-gratis 3D-bildebehandling av personlige lymfocytter. I motsetning til konvensjonelle fluorescens mikroskopi teknikker benytter QPI brytningsindeks (RI) distribusjoner (innebygde optiske egenskaper av levende celler og vev) optisk kontrast6,7. RI tomograms av personlige lymfocytter representerer fenotypiske informasjon spesifikk for subtyper av lymfocytter. I dette tilfellet å systematisk bruke 3D RI tomograms av personlige lymfocytter, ble en veiledet maskin læring algoritmen benyttet.
Bruke ulike QPI teknikker, 3D RI tomograms celler er aktivt brukt for studiet av cellen patofysiologi fordi de gir en etikett-fri, kvantitativ imaging evne8,9,10, 11,12,13. 3D RI distribusjonen av enkeltceller kan også gi morfologiske biokjemiske og biomekaniske informasjon om celler. 3D RI tomograms har tidligere blitt utnyttet innen hematologi14,15,16,17, infeksjonssykdommer18,19, 20, immunologi21, cell biologi22,23, betennelse24, kreft25, nevrovitenskap26,27, utviklingsbiologi28, toksikologi 29og mikrobiologi12,30,31,32.
Selv om 3D RI tomograms gir detaljert morfologiske og biokjemiske informasjon av celler, er klassifiseringen av lymfocytt subtyper vanskelig å oppnå av bare imaging 3D RI tomograms5. Systematisk og kvantitativt utnytte den målte 3D RI tomograms for celle typeklassifisering, benyttet vi en maskin læring algoritme. Nylig har flere blitt rapportert i som kvantitative fase bilder av celler ble analysert med ulike maskin læring algoritmer33, inkludert påvisning av mikroorganismer34, klassifisering av bakterie-slekt35 , 36, rask og etikett-fri påvisning av anthrax sporer37, automatisk analyse av sædceller38, analyse av kreft celler39,40, og oppdagelsen av macrophage aktivisering41.
Denne protokollen gir detaljert fremgangsmåte for å utføre etikett-fri identifikasjon av lymfocytt typer enkeltcelle nivået med 3D QPI og maskinlæring. Dette inkluderer: 1) lymfocytt isolert fra mus blod, 2) lymfocytt sortering via flyt cytometri, 3) 3D QPI, 4) kvantitative egenskapsuttrekking fra 3D RI tomograms og 5) tilsyn læring for å identifisere lymfocytt typer.
Vi presenterer en protokoll som etikett-fri identifikasjon av lymfocytt utnytte 3D kvantitative fase bildebehandling og maskinlæring. Avgjørende skritt i denne protokollen er kvantitative fase bildebehandling og funksjonen utvalg. For optimal holografiske avbilding, bør tetthet av celler kontrolleres som beskrevet ovenfor. Mekanisk stabilitet av cellene er også viktig å få en nøyaktig 3D RI distribusjon fordi flytende eller vibrasjonsmedisin mobilnettet bevegelser vil forstyrre hologrammet målinger på belysning …
