Dit proctocol heeft tot doel een methode te bieden voor in vitro en in vivo mitochondriale Ca2+ beeldvorming in astrocyten en neuronen.
Mitochondriaal Ca2+ speelt een cruciale rol bij het beheersen van cytosolische Ca2+ buffering, energiemetabolisme en cellulaire signaaltransductie. Overbelasting van mitochondriaal Ca2+ draagt bij aan verschillende pathologische aandoeningen, waaronder neurodegeneratie en apoptotische celdood bij neurologische aandoeningen. Hier presenteren we een celtype specifieke en mitochondria gerichte moleculaire benadering voor mitochondriale Ca2 + beeldvorming in astrocyten en neuronen in vitro en in vivo. We construeerden DNA-plasmiden die coderen voor mitochondria-targeting genetisch gecodeerde Ca2 + indicatoren (GECI’s) GCaMP5G of GCaMP6s (GCaMP5G / 6s) met astrocyten- en neuron-specifieke promotors gfaABC1D en CaMKII en mitochondria-targeting sequence (mito-). Voor in vitro mitochondriale Ca2+ beeldvorming werden de plasmiden getransfecteerd in gekweekte astrocyten en neuronen om GCaMP5G/6s tot expressie te brengen. Voor in vivo mitochondriale Ca2+ beeldvorming werden adeno-geassocieerde virale vectoren (AAV’s) voorbereid en geïnjecteerd in de muizenhersenen om GCaMP5G/6s in mitochondriën in astrocyten en neuronen tot expressie te brengen. Onze aanpak biedt een nuttig middel om mitochondriale Ca2 + -dynamiek in astrocyten en neuronen in beeld te brengen om de relatie tussen cytosolische en mitochondriale Ca2 + -signalering te bestuderen, evenals astrocyten-neuroninteracties.
Mitochondriën zijn dynamische subcellulaire organellen en worden beschouwd als de celkrachtcentrales voor energieproductie. Aan de andere kant kunnen mitochondriën Ca2+ opnemen in de matrix als reactie op lokale of cytosolische Ca2+ stijgingen. Mitochondriale Ca2+ opname beïnvloedt de mitochondriale functie, inclusief metabole processen zoals reacties in de tricarbonzuur (TCA) cyclus en oxidatieve fosforylering, en reguleert Ca2+-gevoelige eiwitten onder fysiologische omstandigheden1,2,3,4. Mitochondriale Ca2+ overbelasting is ook een determinant voor celdood, waaronder necrose en apoptose bij verschillende hersenaandoeningen5,6,7. Het veroorzaakt de opening van mitochondriale permeabiliteitstransitieporiën (mPTPs) en de afgifte van caspase-cofactor, die apoptotische celdood initiëren. Daarom is het belangrijk om mitochondriale Ca2 + dynamiek en behandeling in levende cellen te bestuderen om cellulaire fysiologie en pathologie beter te begrijpen.
Mitochondriën handhaven matrix Ca2+ homeostase door een balans tussen Ca2+ opname en efflux. Mitochondriale Ca2+ opname wordt voornamelijk gemedieerd door mitochondriale Ca2+ uniporters (MCU’s), terwijl mitochondriale Ca2+ efflux wordt gemedieerd door de Na+-Ca2+-Li+ wisselaars (NCLXs) en de H+/ Ca2+ wisselaars (mHCXs)8. Het evenwicht kan verstoord worden door de stimulatie van G-eiwit gekoppelde receptoren (GPCR’s)9. Mitochondriale Ca2+ homeostase wordt ook beïnvloed door mitochondriale buffering door de vorming van onoplosbare xCa2+-xPO4x-xOH complexen8.
Intracellulaire en mitochondriale veranderingen in ca2+ concentratie ([Ca2+]) kunnen worden geëvalueerd door fluorescerende of luminescerende Ca2+ indicatoren. Ca2+ binding aan indicatoren veroorzaakt spectrale modificaties, waardoor vrije cellulaire [Ca2+] in real-time in levende cellen kan worden vastgelegd. Er zijn momenteel twee soorten sondes beschikbaar om Ca2+ veranderingen in cellen te monitoren: organische chemische indicatoren en genetisch gecodeerde Ca2+ indicatoren (GECI’s). Over het algemeen zijn verschillende varianten met verschillende Ca2+ affiniteiten (gebaseerd op Kd),spectrale eigenschappen (excitatie- en emissiegolflengten), dynamische bereiken en gevoeligheden beschikbaar voor de onderzochte biologische vragen. Hoewel veel synthetische organische Ca2 + -indicatoren zijn gebruikt voor cytosolische Ca2 + -beeldvorming, kunnen slechts een paar selectief worden geladen in de mitochondriale matrix voor mitochondriale Ca2 + -beeldvorming, waarbij Rhod-2 het meest wordt gebruikt (voor beoordelingen zie10,11). Rhod-2 heeft echter een groot nadeel van lekkage tijdens lange tijd-cursus experimenten; bovendien is het verdeeld tussen mitochondriën, andere organellen en het cytosol, waardoor absolute metingen in verschillende subsecties moeilijk zijn. Door gebruik te maken van celtypespecifieke promotors en subcellulaire compartimentgerichte sequenties, kunnen GECI’s daarentegen worden uitgedrukt in verschillende celtypen en subcellulaire compartimenten voor cel- en compartimentspecifieke Ca2+ beeldvorming in vitro of in vivo. Op fluorescentie-intensiteit gebaseerde GCaMP Ca2+-indicatoren met één golflengte zijn onlangs naar voren gekomen als belangrijke GECI’s12,13,14,15,16. In dit artikel bieden we een protocol voor mitochondria-targeting en celtype specifieke expressie van GCaMP5G en GCaMP6s (GCaMP5G / 6s) in astrocyten en neuronen, en beeldvorming mitochondriale Ca2 + opname in deze celtypen. Met behulp van dit protocol kan de expressie van GCaMP6G / 6s in individuele mitochondriën worden onthuld en kan Ca2 + opname in enkele mitochondriale resolutie worden bereikt in astrocyten en neuronen in vitro en in vivo.
