首先设置自动染色机并准备所有必要的试剂。选择 Daco 程序进行双重染色。输入载玻片的数量和每张载玻片将接受的处理。
根据制造商的建议，将所有试剂加载到自动染色机中。接下来，准备 TBST 和柠檬酸盐、缓冲液、抗体和程序所需的其他试剂。将载玻片放入二甲苯中 5 分钟，以分离载玻片。
不要让载玻片变干，因为这会导致载玻片在去除时出现过多的背景和毛孔染色。将柠檬酸盐缓冲液倒入蒸锅中预热后再使用。接下来，将 depa 玻片提交进行一系列乙醇洗涤。
