标记 M 型均质管并在冰上冷却。标记两个 ml 螺口盖管，并向每个管中加入大约 300 μL 的氧化锆珠。准备好以下仪器以供使用，包括组织解离器、微珠打浆器、带有 50 毫升试管和 96 孔板适配器的台式离心机以及光度计。
从 80 摄氏度的冰箱中取出从白色念珠菌感染的小鼠中分离的先前冷冻的肾脏，并放在冰上。向每个肾脏中加入 1.2 毫升缓冲液 RLT。然后将每个装有缓冲液的肾脏倒入 M 型匀浆管中。
使用的缓冲区 RLT 量是根据经验确定的。缓冲液 RLT 过多会稀释样品浓度，而缓冲液 RLT 过多会使匀浆非常粘稠且极难处理。接下来，在预装的 RNA 2.01 上使用组织解离剂，匀浆肾脏。

要使用数字条形码进行基因表达分析，首先在冰上解冻报告基因代码集和捕获代码集。向报告基因代码集中添加 130 μL 杂交缓冲液。Invert 进行混合并向下旋转。
接下来，向 12 个反应管中的每个反应管中加入 20 微升混合物。然后向每管中加入 10 μg 总组织 RNA，并通过移液混合。根据感染模型、接种量和使用的病原体菌株，病原体 RNA 在总 RNA 中的百分比从 0 到 2% 不等，因此应根据经验优化需要为每种添加的 RNA 总量。
现在向每个试管中添加 5 微升捕获代码组。通过翻转试管混合并快速旋转。然后在 65 摄氏度的热循环仪中用加热盖孵育反应物约 18 小时。
从 20 摄氏度中取出一个样品柱，使其升温至室温。从 4 摄氏度中取出两个试剂板，在台式离心机中以 670 x G 离心 2 分钟。接下来，按照屏幕上的分步说明设置制备站，选择高灵敏度选项。
然后从热循环仪中取出反应物，并立即加载到制备站上。运行三个小时的高灵敏度程序后，取出小柱并使用透明胶带密封泳道。如果需要，将矿物油涂抹在墨盒底部。
然后将小柱加载到数字分析仪上。按照屏幕上的分步说明设置数字分析仪，选择高分辨率扫描选项。运行扫描程序后，下载结果或选择通过电子邮件接收结果，并将原始数据导入制造商的软件。
