大约在第 14 至 21 天，出现明显的跨分化区域。这些原代菌落缺乏典型的滋养层干细胞形态，但是一旦它们被传代培养，就会观察到具有紧密边缘和明亮边界的特征性上皮形态，很容易让人想起真正的滋养层干细胞。当这四个因子诱导的滋养层干细胞对滋养层转录因子、 Cdx2 和 Tfap2c 保持阳性时，这些细胞系的想法可用于后续的体外或体内分析。
