24、48 和 96 小时后，用 1 毫升新鲜的完全培养基替换每个插件顶部的培养基。每个孔底部的培养基中加入 2 毫升新鲜完全培养基。共培养 120 小时后，将一个插入片段一次转移到含有 1 mL PBS 的单个 35 mm 细胞培养皿中。
并用 1 毫升 PBS 清洗插入物的底部和顶部。要收集在插入片段底部生长的细胞，请将插入片段底部朝下放入 200 μL 室温胰蛋白酶-EDTA 中。在室温下放置 2 分钟后，用 800 μL 完全生长培养基终止反应。

第三次洗涤后，将细胞在室温下用 4% 甲醛的 PBS 溶液固定 10 分钟，然后用 Tris 缓冲盐水洗涤五次。最后一次洗涤后，用添加 0.1 皂苷的 0.25% Triton X-100 在室温下透化细胞 5 分钟，然后在封闭溶液中在室温下孵育 1 小时。接下来，用感兴趣的一抗标记样品，在 4 摄氏度下封闭过夜。

0.4 微米孔插入片段还限制了插入片段两侧细胞培养物之间功能性间隙连接的形成，从而限制了与分泌因子的通讯。然而，具有 1 微米和 3 微米孔的插入片段允许细胞通过间隙连接进行功能偶联，如在这些共培养物中荧光标记跨膜转移所示。重要的是，该系统可用于在正常人二倍体成纤维细胞与乳腺癌细胞共培养后有效地从它们产生癌症相关的成纤维细胞，与人乳腺癌细胞共培养的成纤维细胞上 caveolin-1 的表达降低证明了这一点。
