接下来，离心管以沉淀细胞。然后，小心吸出并丢弃所得上清液。重悬沉淀，使用 1 mL PBS 制备单细胞悬液。
然后，短暂离心试管。再次沉淀并重悬细胞，以完全去除培养基。接下来，将细胞沉淀重悬于 600 微升 PBS 中。

为了估计总蛋白的细胞内表达，接下来标记透化细胞。这是使用与细胞外标记类似的方案完成的，但添加了基于皂苷的细胞透化缓冲液。沉淀细胞，弃去上清液，直接从制备的原液中将细胞重悬于 100 微升固定透化溶液中。
将细胞在 4 摄氏度的黑暗中孵育 20 分钟。然后，加入 100 微升新鲜制备的透化洗涤缓冲液，并短暂涡旋细胞。接下来，沉淀细胞并使用透化洗涤缓冲液再洗涤两次。
然后，加入 5 μg/mL 的 FITC 偶联单克隆抗 HA 抗体和 100 μL 透化洗涤缓冲液。短暂涡旋细胞，并在 4 摄氏度的黑暗中孵育细胞 30 分钟。接下来，加入 100 微升透化洗涤缓冲液。
沉淀细胞并在 100 μL 相同缓冲液中洗涤 3 次，如前所述。最后一次洗涤后，将细胞重悬于 500 μL PBS 中，并将单细胞悬液转移到 5 mL 流式细胞仪管中。在同一天通过流式细胞仪运行未透化和透化的细胞。
