Beskrivning av en kvantitativ fosforylering förfarande med cryolysis, urea solubilziation, HILIC fraktionering och iMac anrikning av fosforylerade peptider.
Detta protokoll beskriver tillväxt och stimulans, med fettsyror oleat av isotopiskt tunga och lätta S. cerevisiae celler. Celler mals med hjälp av en cryolysis förfarande i en boll kvarn kvarn och de resulterande grindate bringas i lösning genom att urea lösningsgörande. Detta förfarande gör det möjligt att lys av cellerna i ett metaboliskt inaktiva tillstånd, bevara fosforylering och förebygga omorientering av phosphoproteome under cellslys. Efter sönderdelning alkylering, trypsin nedbrytning av de proteiner, är proverna desalted på C18 kolonner och provet komplexiteten minskas genom fraktionering med hydrofil interaktion kromatografi (HILIC). HILIC kolumner behålla företrädesvis hydrofila molekyler som är väl lämpad för phosphoproteomics. Fosforyleras peptider brukar eluera senare i kromatografiska profil än den icke fosforyleras motsvarigheter. Efter fraktionering är phosphopeptides berikad med immobiliserade metall kromatografi, som bygger på laddning-baserade affinitet för phosphopeptide anrikning. I slutet av detta förfarande proverna är klara för kvantitativt analyseras med masspektrometri.
Denna metod kommer att ge en anrikning av phosphopeptides som kan kvantitativt analyseras. Ett antal parametrar kan ändras i detta protokoll, men det viktigaste att komma ihåg är att bevara din fosforylering. Beredning av celler för kvantifiering kan uppnås på flera olika sätt, till exempel om du inte kan märka dina celler in vivo, taggar såsom ICAT [3] eller ITRAC [4] kan användas efter utvinning till differentiellt etikett två kulturer för kvantifiering syften.
