Een methode van het gebruik van solid-state nanoporiën om de niet-specifieke adsorptie van eiwitten te controleren op een anorganisch oppervlak is beschreven. De methode maakt gebruik van de resistieve-puls principe, waardoor de adsorptie aan gezocht worden in real-time en op de single-molecule-niveau. Omdat het proces van enkel eiwit-adsorptie wordt ver van evenwicht, stellen wij de werkgelegenheid van parallelle arrays van synthetische nanoporiën, waardoor voor de kwantitatieve bepaling van de schijnbare eerste orde reactie snelheidsconstante van eiwitadsorptie evenals en de Langmuir adsorptie constant.
Solid-state nanoporiën zijn gebruikt om metingen uit te voeren op de single-molecule niveau naar het lokale structuur en flexibiliteit van nucleïnezuren 1-6, het ontvouwen van eiwitten 7, en de bindingsaffiniteit van verschillende liganden acht te onderzoeken. Door het koppelen van deze nanoporiën om de resistieve-pulstechniek 9-12, kan dergelijke metingen worden gedaan onder een breed scala van omstandigheden en zonder de noodzaak voor etikettering 3. In de resistieve-pulse-techniek, is een ionisch zout-oplossing geïntroduceerd op beide zijden van de nanogaatje. Daarom zijn ionen verdreven uit de ene kant van de kamer naar de andere door een toegepaste transmembraan potentieel, wat resulteert in een constante stroom. Het partitioneren van een analyt in de nanogaatje veroorzaakt een goed gedefinieerde doorbuiging in de huidige, die kan worden geanalyseerd om single-molecule informatie eruit te halen. Met behulp van deze techniek kan de adsorptie van enkele eiwitten aan de nanogaatje muren worden gecontroleerd onder een breed scala aanvoorwaarden 13. Eiwit-adsorptie wordt steeds belangrijker, want als microfluïdische apparaten krimpen in omvang, de interactie van deze systemen met enkele eiwitten wordt een punt van zorg. Dit protocol beschrijft een snelle test voor eiwit binding aan nitride films, die gemakkelijk kan worden uitgebreid tot andere dunne films vatbaar voor nanogaatje boren, of om gefunctionaliseerde oppervlakken nitride. Een verscheidenheid van eiwitten kunnen worden bestudeerd onder een breed scala aan oplossingen en denaturerende omstandigheden. Bovendien kan dit protocol worden gebruikt om meer fundamentele problemen met behulp van spectroscopie nanogaatje te verkennen.
Spontane adsorptie van eiwitten op solid-state oppervlakken 27-29 is van fundamenteel belang in een aantal gebieden, zoals de biochip toepassingen en het ontwerp van een nieuwe klasse van functionele hybride biomaterialen. Eerdere studies hebben aangetoond dat eiwitten geadsorbeerd aan solid-state oppervlakken geen laterale mobiliteit of significante desorptie-tarief is gebleken, en daarom eiwit-adsorptie wordt algemeen beschouwd als een onomkeerbaar en niet-specifieke proces van 30-32.
