Eine Methode der Verwendung von Solid-State-Nanoporen, die unspezifische Adsorption von Proteinen auf einer anorganischen Oberfläche überwachen beschrieben. Da der Prozess der einzelnen Protein-Adsorption ist fern vom Gleichgewicht, schlagen wir den Einsatz von parallel Arrays von synthetischen Nanoporen, so dass für die quantitative Bestimmung der scheinbaren Reaktion erster Ordnung Geschwindigkeitskonstante der Proteinadsorption sowie die Langmuir Adsorption konstant.
Solid-State-Nanoporen wurden genutzt, um Messungen an der Einzel-Molekül-Ebene durchzuführen, um die lokale Struktur und Flexibilität von Nukleinsäuren 1-6, die Entfaltung von Proteinen 7 und Bindungsaffinität unterschiedlicher Liganden 8 zu untersuchen. Durch die Kopplung dieser Nanoporen der resistiven-Puls-Technik 9-12, können solche Messungen unter einer Vielzahl von Bedingungen und ohne die Notwendigkeit für die Kennzeichnung 3 durchgeführt werden. In der resistive-Puls-Technik ist ein ionisches Salz-Lösung auf beiden Seiten des Nanopore eingeführt. Daher werden die Ionen von einer Seite der Kammer in die andere durch eine angelegte Transmembranpotential getrieben, was zu einem stetigen Strom. Die Aufteilung eines Analyten in die Nanopore Ursachen einer gut definierten Auslenkung in diesem Strom, der analysiert, um Einzel-Molekül-Informationen zu extrahieren können.
