Xenopus laevis gir en ideell modell system for å studere cellen skjebne spesifikasjon og fysiologiske funksjon av individuelle netthinnens celler i grunnskolen cellekultur. Her presenterer vi en teknikk for å dissekere retinal vev og generere primære cellekulturer som er avbildet for kalsium aktivitet og analysert ved in situ hybridisering.
Den prosessen som den fremre delen av nevrale plate gir opphav til virveldyr netthinnen fortsetter å være et stort fokus på både klinisk og grunnleggende forskning. I tillegg til de åpenbare medisinsk relevans for å forstå og behandle retinal sykdom, fortsetter utviklingen av virveldyr netthinnen å tjene som et viktig og elegant modellsystem for forståelse neuronal celletype bestemmelse og differensiering 1-16. Det nevrale netthinnen består av seks diskrete celletyper (ganglion, amacrine, horisontal, fotoreseptorer, bipolare celler, og Muller gliacellene) ordnet i stereotype lag, et mønster som er i stor grad bevart blant alle virveldyr 12,14-18.
Mens han studerte netthinnen i intakt utvikling av embryo er klart nødvendig for å forstå hvordan dette komplekse organ utvikler seg fra en utvekst av forebrain inn en lagdelt struktur, er det mange spørsmål som er til fordel from ansette tilnærminger med primær cellekultur av presumptive netthinnens celler 7,19-23. For eksempel, å analysere celler fra vev fjernet og dissosiert på forskjellige stadier tillater en å skjelne staten spesifikasjonen av individuelle celler på ulike utviklingsstadier, dvs. skjebnen av cellene i fravær av interaksjoner med nærliggende vev 8,19-22 ,24-33. Primære cellekulturer kan også etterforsker for å behandle kultur med spesifikke reagenser og analysere resultatene på en enkelt celle nivå 5,8,21,24,27-30,33-39. Xenopus laevis, en klassisk modell system for studiet av tidlig nevral utvikling 19,27,29,31-32,40-42, fungerer som en spesielt egnet system for retinal primære cellekulturer 10,38,43-45.
Presumptive retinal vev er tilgjengelig fra de tidligste stadiene av utviklingen, umiddelbart etter nevrale induksjon 25,38,43. I tillegg gis thved hver celle i embryoet inneholder en tilførsel av eggeplomme, kan netthinnens celler dyrkes i en svært enkel definerte medier bestående av en bufret salt-løsning, og dermed fjerne forstyrrende effekter av inkubering eller andre sera-baserte produkter 10,24,44-45 .
Imidlertid er isolasjon av retinal vev fra omkringliggende vev og påfølgende behandling utfordrende. Her presenterer vi en fremgangsmåte for disseksjon og dissosiasjon av netthinnens celler i Xenopus laevis som skal anvendes for å fremstille primære cellekulturer som vil, i sin tur, blir analysert for kalsium aktivitet og genekspresjon på oppløsningen av enkeltceller. Mens temaet presenteres i denne artikkelen er analysen av spontane kalsium transienter, er teknikken bredt gjeldende for et bredt spekter av problemstillinger og tilnærminger (figur 1).
Med sine godt karakterisert celletyper som er bevart på tvers av alle virveldyr, gir netthinnen en nyttig modell for å studere molekylære cellulære prosesser som styrer celletype spesifikasjon og differensiering.
