Xenopus laevis ger en idealisk modell för att studera specifikation cellöde och fysiologisk funktion hos individuella retinala celler i primär cellkultur. Här presenterar vi en teknik för dissekera retinal vävnad och generera primära cellkulturer som avbildas för kalcium aktivitet och analyserades genom in situ hybridisering.
Den process genom vilken den främre regionen av neurala plattan ger upphov till ryggradsdjur näthinnan fortsätter att vara ett stort fokus på både klinisk forskning och grundforskning. Förutom den uppenbara medicinska relevans för att förstå och behandla näthinnesjukdom, fortsätter utvecklingen av ryggradsdjur näthinnan att fungera som ett viktigt och elegant modell för att förstå neuronala celltyp bestämning och differentiering 1-16. Den neurala näthinnan består av sex separata celltyper (ganglion, amacrine, horisontella, fotoreceptorer, bipolära celler och Muller gliaceller) arrangerade i stereotypa lager, ett mönster som till stor del är bevarad bland alla ryggradsdjur 12,14-18.
Medan de studerar näthinnan i intakt utvecklande embryot är klart krävs för att förstå hur denna komplexa organ utvecklas från ett utsprång av framhjärnan till en skiktad struktur, det finns många frågor som gynnar tillbakam anställa metoder som använder primär cellkultur av presumtiva retinala celler 7,19-23. Till exempel, analysera celler från vävnader avlägsnats och dissocierad i olika skeden gör att man kan urskilja tillstånd specificering av individuella celler vid olika utvecklingsstadier, dvs öde cellerna i frånvaro av interaktioner med närliggande vävnader 8,19-22 ,24-33. Primär cellodling ger också utredaren att behandla kulturen med specifika reagens och analysera resultaten på en enda cell nivå 5,8,21,24,27-30,33-39. Xenopus laevis, ett klassiskt modellsystem för att studera tidig neural utveckling 19,27,29,31-32,40-42, fungerar som en särskilt lämplig för retinal primär cellkultur 10,38,43-45.
Presumtiv näthinnevävnad är tillgänglig från de tidigaste utvecklingsstadierna, omedelbart efter neural induktion 25,38,43. Med tanke på thvid varje cell i embryot innehåller ett förråd av äggula, kan retinala celler odlas i ett definierat mycket enkel medier bestående av en buffrad saltlösning och därmed undanröja de förbryllande effekterna av inkubering eller andra serum-baserade produkter 10,24,44-45 .
Emellertid är isoleringen av den retinala vävnaden från omgivande vävnader och den efterföljande bearbetningen utmanande. Här presenterar vi en metod för dissektion och dissociation av näthinneceller i Xenopus laevis som kommer att användas för att framställa primära cellkulturer som, i sin tur, kan analyseras för kalcium aktivitet och genexpression vid upplösningen av enskilda celler. Medan ämnet som presenteras i denna uppsats är en analys av spontana kalcium transienter är tekniken brett tillämpbar för ett brett spektrum av frågeställningar och metoder (figur 1).
Med sina välkarakteriserade celltyper som är konserverade i alla ryggradsdjur, ger näthinnan en användbar modell för att studera molekylär cellulära processer som styr specifikation celltyp och differentiering. Primär cellkultur ger en kraftfull metod för att undersöka ett stort antal processer, inklusive genexpression, dynamik protein och kalcium och elektrisk aktivitet vid nivån för en enda cell upplösning.
