Kolonisering af murine nasopharynx med Streptococcus pneumoniae og den efterfølgende ekstraktion af vedhængende eller rekrutterede celler er beskrevet. Denne teknik indebærer skylning af nasopharynx og indsamling af væsken gennem nares og kan tilpasses til forskellige udlæsninger, herunder differentialcellekvantificering og analyse af mRNA-udtryk in situ.
Nasopharyngeal kolonisering af Streptococcus pneumoniae er en forudsætning for invasion i lungerne eller blodbanen1. Denne organisme er i stand til at kolonisere nasopharynxens slimhindeoverflade, hvor den kan opholde sig, formere sig og til sidst overvinde værtsforsvaret for at invadere til andre væv af værten. Etablering af en infektion i de normalt lavere luftveje resulterer i lungebetændelse. Alternativt kan bakterierne spredes i blodbanen, der forårsager bakterieæmi, som er forbundet med højdødelighed 2, ellers fører direkte til udvikling af pneumokok meningitis. Forståelse af kinetik af, og immunrespons på, nasopharyngeal kolonisering er et vigtigt aspekt af S. lungebetændelse infektion modeller.
Vores musemodel af intranasal kolonisering er tilpasset fra menneskelige modeller3 og er blevet brugt af flere forskergrupper i undersøgelsen af værtspatogenresponser i nasopharynx4-7. I den første del af modellen bruger vi et klinisk isoleret S. pneumoniae til at etablere en selvbegrænsende bakteriel kolonisering, der ligner transporthændelser hos voksne mennesker. Den procedure, der er beskrevet heri, indebærer forberedelse af et bakterielt inoculum efterfulgt af etablering af en koloniseringshændelse gennem levering af inoculumet via en intranasal indgiftsmåde. Residente makrofager er den fremherskende celletype i nasopharynx under konstant tilstand. Typisk er der få lymfocytter til stede i uinficerede mus8, men slimhindekolonisering vil føre til lav- til højkvalitetsbetændelse (afhængigt af virulensen af bakteriearterne og stammen), der vil resultere i et immunrespons og den efterfølgende rekruttering af værtsimmunceller. Disse celler kan isoleres af et toilet af trachealindholdet gennem narerne og korreleres med massefylden af koloniseringsbakterier for bedre at forstå infektionens kinetik.
I denne undersøgelse præsenterede vi detaljerede metoder til intranasal kolonisering af mus ved hjælp af en klinisk isolere stamme af Streptococcus pneumoniae og den efterfølgende isolation og karakterisering af immuncellerne rekrutteret til nasopharynx som reaktion på bakterierne. Vi demonstrerede, hvordan et bakterielt inoculum kan dyrkes i næringsrige medier og bruges til at etablere en koloniseringsbegivenhed hos mus, som oprindeligt er begrænset til nasopharynx.
