Kolonisering av murin nasopharynx med Streptococcus pneumoniae og etterfølgende utvinning av tilhenger eller rekrutterte celler er beskrevet. Denne teknikken innebærer å skylle nasopharynx og samling av væsken gjennom nares og er tilpasningsdyktig for ulike avlesninger, inkludert differensialcelle kvantifisering og analyse av mRNA uttrykk in situ.
Nasopharyngeal kolonisering av Streptococcus pneumoniae er en forutsetning for invasjon til lungene eller blodbanen1. Denne organismen er i stand til å kolonisere slimhinnens overflate av nasopharynx, hvor den kan ligge, formere seg og til slutt overvinne vertsforsvar for å invadere til andre vev av verten. Etablering av en infeksjon i normalt nedre luftveier resulterer i lungebetennelse. Alternativt kan bakteriene spre seg inn i blodet og forårsake bakterieemi, som er forbundet med høy dødelighet2, eller ellers føre direkte til utvikling av pneumokokk meningitt. Å forstå kinetikken til, og immunresponser på, nasofaryngeal kolonisering er et viktig aspekt av S. lungebetennelse infeksjonsmodeller.
Vår musemodell av intranasal kolonisering er tilpasset fra menneskelige modeller3 og har blitt brukt av flere forskningsgrupper i studiet av vertspatogenresponser i nasopharynx4-7. I den første delen av modellen bruker vi en klinisk isolering av S. pneumoniae for å etablere en selvbegrensende bakteriekolonisering som ligner på vognhendelser hos menneskelige voksne. Prosedyren som er beskrevet her, innebærer forberedelse av et bakteriell inokulum, etterfulgt av etableringen av en koloniseringshendelse gjennom levering av inoculum via en intranasal administrasjonsvei. Residente makrofager er den dominerende celletypen i nasopharynx under steady state. Vanligvis er det få lymfocytter tilstede hos uinfiserte mus8, men slimhinnekolonisering vil føre til lav- til høygradig betennelse (avhengig av virulensen av bakterielle arter og belastning) som vil resultere i en immunrespons og den påfølgende rekrutteringen av vertsimmunceller. Disse cellene kan isoleres av en lavage av trakealinnholdet gjennom nares, og korrelert med tettheten av koloniseringsbakterier for bedre å forstå infeksjonens kinetikk.
I denne studien presenterte vi detaljerte metoder for intranasal kolonisering av mus ved hjelp av en klinisk isolerende stamme av Streptococcus pneumoniae og den påfølgende isolasjonen og karakteriseringen av immuncellene rekruttert til nasopharynx som svar på bakteriene. Vi demonstrerte hvordan et bakteriell inokulum kan dyrkes i næringsrike medier og brukes til å etablere en koloniseringshendelse hos mus, som i utgangspunktet er begrenset til nasopharynx.
