Sono stati tradizionalmente condotti studi funzionali del sistema uditivo dei mammiferi utilizzando tecniche spazialmente concentrati come registrazioni elettrofisiologiche. Il seguente protocollo descrive un metodo di visualizzare modelli su larga scala di attività emodinamica evocata nel gatto corteccia uditiva utilizzando la risonanza magnetica funzionale.
Le attuali conoscenze di elaborazione sensoriale nel sistema uditivo dei mammiferi è derivato principalmente da studi elettrofisiologici in una varietà di modelli animali, tra cui scimmie, furetti, pipistrelli, roditori e gatti. Al fine di elaborare adeguate paralleli tra modelli umani e animali della funzione uditiva, è importante stabilire un ponte tra gli studi di imaging funzionale umani e studi elettrofisiologici animali. Risonanza magnetica funzionale (fMRI) è un consolidato metodo minimamente invasivo per misurare vasti reticoli di attività emodinamica nelle diverse regioni della corteccia cerebrale. Questa tecnica è ampiamente utilizzata per sondare la funzione sensoriale nel cervello umano, è uno strumento utile nel collegamento studi di elaborazione uditiva in entrambi gli esseri umani e gli animali ed è stato utilizzato con successo per studiare la funzione uditiva nelle scimmie e roditori. Il protocollo che segue descrive una procedura sperimentale per studiare la funzione uditiva in anestetizzati adultigatti dalla misura di stimolo evocato cambiamenti emodinamici nella corteccia uditiva utilizzando fMRI. Questo metodo agevola il confronto tra le risposte emodinamiche attraverso diversi modelli di funzione uditiva dando luogo ad una migliore comprensione delle caratteristiche specie-indipendente della corteccia uditiva dei mammiferi.
Comprensione attuale di elaborazione uditiva nei mammiferi è derivato principalmente da studi elettrofisiologici invasivi nelle scimmie 1-5, furetti 6-10, 11-14 pipistrelli, roditori 15-19, 20-24 e gatti. Tecniche elettrofisiologiche comunemente utilizzano microelettrodi extracellulari per registrare l'attività dei neuroni singoli e multipli all'interno di una piccola area di tessuto neurale che circonda la punta dell'elettrodo. Fondata metodi di imaging funzionale, come l'imaging ottico e la risonanza magnetica funzionale (fMRI), complementi servire come utili a registrazioni extracellulari, fornendo un punto di vista macroscopico di attività ad azionamento simultaneo su più regioni spazialmente distinte del cervello. Segnale intrinseco imaging ottico facilita la visualizzazione di attività evocata nel cervello misurando i cambiamenti di attività connesse nelle proprietà di riflettanza del tessuto superficie, mentre fMRI utilizza il sangue di ossigeno livello-dipendente (BOLD)contrasto per misurare stimolo evocato alterazioni emodinamiche nelle regioni del cervello che sono attive durante una determinata attività. Imaging ottico richiede l'esposizione diretta della superficie corticale di misura le variazioni di superficie riflettanza tessuto che sono legati all'attività di stimolo evocato 25. In confronto, fMRI è invasiva e sfrutta le proprietà paramagnetiche di sangue deoxygenated di misurare sia la superficie corticale 26-28 e basati solco-27,29 attività evocata all'interno di un cranio intatto. Forti correlazioni tra il segnale BOLD e l'attività neuronale in primati non umani corteccia visiva 30 e nella corteccia uditiva umana 31 convalidano fMRI come uno strumento utile per studiare la funzione sensoriale. Dal fMRI è stato ampiamente utilizzato per studiare le caratteristiche del percorso uditivo quali l'organizzazione tonotopic 32-36, lateralizzazione della funzione uditiva 37, pattern di attivazione corticale, l'identificazione di regioni corticali 38, effetti del suonointensità sulle proprietà di risposta uditive 39,40, e le caratteristiche della risposta BOLD andamento temporale 29,41 in umano, scimmia, e modelli di ratto, lo sviluppo di un protocollo adatto imaging funzionale per studiare la funzione uditiva nel gatto fornirebbe un utile complemento letteratura imaging funzionale. Mentre fMRI è stato utilizzato anche per esplorare i vari aspetti funzionali della corteccia visiva nel gatto anestetizzato 26-28,42, pochi studi hanno utilizzato questa tecnica per esaminare l'elaborazione sensoriale gatto corteccia uditiva. Lo scopo del presente protocollo è di stabilire un metodo efficace di utilizzare fMRI quantificare funzione nella corteccia uditiva del gatto anestetizzato. Le procedure sperimentali descritte in questo manoscritto sono stati utilizzati con successo per descrivere le caratteristiche del corso BOLD tempo di risposta nel gatto adulto corteccia uditiva 43.
Nella progettazione di un esperimento fMRI per un modello anestetizzato della funzione uditiva degli animali, le seguenti questioni dovrebbe essere data attenta considerazione: (i) l'impatto di anestesia sulle risposte corticali, (ii) l'effetto del rumore di fondo dello scanner, e (iii) l'ottimizzazione della fase di raccolta dei dati della procedura sperimentale.
