Twee technieken voor het isoleren van cellulaire lipide druppeltjes uit 1) gistcellen en 2) humane placenta worden gepresenteerd. De kern van beide procedures is dichtheidsgradiëntcentrifugatie, waar de resulterende drijvende laag met de druppeltjes gemakkelijk kan worden gevisualiseerd door oog, gewonnen, en gekwantificeerd door Western Blot analyse voor zuiverheid.
Lipidedruppeltjes zijn dynamische organellen die gevonden kunnen worden in de meeste eukaryote en bepaalde prokaryote cellen. Structureel, de druppels uit een kern van neutrale lipiden omringd door een fosfolipide monolaag. Een van de meest nuttige technieken het bepalen van de cellulaire rol van druppels is proteomische identificatie van gebonden eiwitten, die kunnen worden geïsoleerd met de druppels. Hier worden twee methoden beschreven om lipide druppels en hun gebonden eiwitten te isoleren van twee brede eukaryoten: splijtgist en menselijke placenta villous cellen. Hoewel beide technieken verschillen, de belangrijkste methode – dichtheidsgradiëntcentrifugatie – wordt gedeeld door beide preparaten. Dit toont de brede toepasbaarheid van de gepresenteerde druppel isolatietechnieken.
In het eerste protocol, worden gistcellen omgezet in sferoplasten door enzymatische afbraak van de celwanden. De resulterende sferoplasten worden vervolgens gently gelyseerd in een loszittende homogenisator. Ficoll wordt toegevoegd aan het lysaat aan een dichtheidsgradiënt geven en het mengsel wordt driemaal gecentrifugeerd. Na de eerste rotatie, worden de lipidedruppeltjes gelokaliseerd in het witgekleurde drijflaag van de centrifugebuizen met het endoplasmatisch reticulum (ER), de plasmamembraan en vacuolen. Twee daaropvolgende spins worden gebruikt om deze drie andere organellen verwijderen. Het resultaat is een laag die alleen druppels en gebonden eiwitten heeft.
In het tweede protocol, worden placenta villous cellen geïsoleerd uit menselijke term placenta door inwerking van het enzym trypsine en DNase I. De cellen worden gehomogeniseerd in een loszittende homogenisator. Lage snelheid en gemiddelde snelheid centrifugeren stappen worden gebruikt om gebroken cellen, cellulaire puin, kernen, en mitochondriën verwijderen. Sucrose wordt aan het homogenaat tot een dichtheidsgradiënt geven en het mengsel wordt gecentrifugeerd om de lipide druppels onderscheiden van andere cellular fracties.
De zuiverheid van het lipide druppels in beide protocollen wordt bevestigd door Western Blot analyse. De druppel fracties uit beide desinfectantia zijn geschikt voor latere proteomics en lipidomic analyse.
Cellulaire lipide druppels zijn dynamische organellen die meerdere functies dienen in cellen. Ze zijn opslag knooppunten voor neutrale lipiden, die kan worden omgezet in energie of voor fosfolipide synthese. De druppels spelen een centrale rol in fysiologische en pathologische omstandigheden waaronder atherosclerose, obesitas en gerelateerde metabole ziekten, en ook infectieziekten 1,2. Bovendien, ze zijn intrigerend bronnen voor biodiesel.
Veel informatie over de cellulaire rol van lipide druppels is verkregen van proteomics en lipidomic analyse van druppeltjes gezuiverd uit uiteenlopende organismen 3. Deze organismen hebben opgenomen bacteriën 4,5, gist 6-11, planten 12,13, aaltjes 14, en vliegt 15,16. Gezien het belang van de rol van lipide druppels in menselijke stofwisselingsziekten, druppeltjes zijn ook geïsoleerd uit gekweekte dierlijke cellen en eenNimal weefsels. Gekweekte cellijnen 3T3-L1 adipocyten 17, Chinese hamster ovarium (CHO)-cellen K2 18, humane hepatocyten 19,20 en epitheliale cellijnen 21 opgenomen. Dierlijke weefsels waaruit druppeltjes zijn geïsoleerd zijn muis skeletspieren 22, 23 lever en borstklieren 23 inbegrepen. Zoals hierboven vermeld, het doel van de meeste druppel isolatie studies is proteoomanalyse voeren op de afhankelijke factoren en lipiden analyse van de neutrale en fosfolipiden.
Sinds neutrale lipiden – de meest talrijke onderdeel van lipide druppels – zijn minder dicht dan de meeste andere cellulaire materialen, heeft isolatie van druppeltjes traditioneel met dichtheidsgradiëntcentrifugatie. Die techniek is het middelpunt van beide desinfectantia hier gepresenteerd. Vorige technieken 6,24 worden gecombineerd en bewerkt tot een visuele presentatie van de isolatie van druppeltjes uit gekweekte kernsplijting gistcellenen noncultured menselijke cellen verkregen van placenta weefsel. Het doel is de brede toepasbaarheid van deze techniek zien door te kiezen twee sterk verschillende celtypes als uitgangspunten voor de druppel isolatie. Deze techniek moet nuttig zijn voor diegenen die druppels van de meeste organismen te isoleren.
Protocol 1 beschrijft de isolatie van lipide druppeltjes uit de splijtgist, Schizosaccharomyces pombe, die is gebruikt als een model voor het waarnemen druppelvorming in eukaryote celdeling 25. De gist Saccharomyces cerevisiae is uitgebreid gebruikt als modelorganisme voor het bestuderen van lipide druppel biologie. Protocol nr. 1 is van toepassing op zowel organismen en verschillen in de voorbereidingen worden gemarkeerd.
Protocol 2 beschrijft de isolatie van lipide druppeltjes uit placenta villous cellen, die op hun beurt verkregen uit menselijk term placenta. Decollectie term placenta's biedt een unieke gelegenheid om 200-250 g beschikbaar menselijk weefsel 26, die aanzienlijke aantallen lipide druppels bevat veilig en ethisch te verkrijgen. Dit in tegenstelling tot de meeste lipide druppel isolatie werken in hogere eukaryoten waar de druppels afkomstig uit gekweekte cellen. In deze studies worden vetzuren vaak toegevoegd aan de kweek tot de synthese van neutrale lipiden en daarmee de groei van druppeltjes te bevorderen. Dit in tegenstelling tot de werk hier waar lipide druppeltjes worden gevormd onder natuurlijke omstandigheden in placentaweefsel.
De zuiverheden van het lipide druppel fracties worden bepaald door Western blot analyse onder toepassing organel marker antilichamen. Deze twee protocollen lipide druppel fracties die geschikt zijn voor daaropvolgende proteomische en lipiden analyse opleveren.
Kritische stappen binnen dit protocol
Zorg overeenstemming met media en celdichtheden te zijn bij de groei van gekweekte cellen. Cellular lipidedruppeltjes zijn uniek omdat hun geassocieerde eiwitten zijn sterk afhankelijk van de omgeving waarin de cellen worden gekweekt 17. Daarom moet het medium waarin de cellen worden gekweekt en de dichtheid van de cellen nauwkeurig gecontroleerd vóór lyse.
