A protocol is described to extract primary human cells from surgical colon tumor and normal tissues. The isolated cells are then cultured on soft elastic substrates (polyacrylamide hydrogels) functionalized by an extracellular matrix protein, and embedded with fluorescent microbeads. Traction cytometry is performed to assess cellular contractile stresses.
Kankercellen reageren op mechanische stijfheid matrix op een complexe wijze met een gecoördineerde hiërarchische mechanisch-chemisch systeem bestaande uit adhesiereceptoren en geassocieerde signaaltransductie membraaneiwitten, het cytoskelet architectuur en moleculaire motors 1, 2. Mechanische gevoeligheid van verschillende kankercellen in vitro voornamelijk onderzocht met onsterfelijk gemaakte cellijnen of primaire cellen van muizen afgeleide, niet met primaire humane kankercellen. Derhalve is er weinig bekend over de mechanische gevoeligheid van primaire humane colon kankercellen in vitro. Hier, wordt een geoptimaliseerd protocol ontwikkeld die de isolatie van primaire humane colon van gezonde cellen en kankercellen chirurgische menselijke weefselmonsters beschreven. Geïsoleerd colon cellen worden vervolgens met succes gekweekt op zachte (2 kPa stijfheid) en stijf (10 kPa stijfheid) polyacrylamide hydrogels en stijve polystyreen (~ 3,6 GPa stijfheid) substraten gefunctionaliseerd door een extracellulaire matrix (fibronectinein dit geval). Fluorescerende microbolletjes zijn ingebed in zachte gels in de buurt van de celkweek oppervlak, en tractie test wordt uitgevoerd om cellulaire samentrekkende spanningen met behulp van gratis open toegang software te beoordelen. Bovendien immunofluorescentie microscopie van verschillende stijfheid substraten nuttige informatie over primaire celmorfologie, cytoskelet organisatie en vinculin met focale adhesies als functie van substraat stijfheid.
De laatste jaren is het steeds duidelijker geworden dat de mechanische micro-omgeving, naast biochemische factoren, speelt een belangrijke rol bij het reguleren cel functionaliteiten. Cellen kunnen detecteren en reageren op de stijfheid substraat waarop zij worden nageleefd (zoals in 2D kweek) en omringd door (zoals in 3D kweek) 3-7. Hierdoor kunnen cellen hun differentiatie 3, morfologie 4, migratie / motiliteit 5, biofysische eigenschappen 6, 7 groei, en andere processen moduleren.
Kankercellen ook reageren op de 2D- en 3D-matrix stijfheid met behulp van een gecoördineerde, hiërarchische mechanisch-chemische combinatie van adhesie receptoren en bijbehorende signaaltransductie membraaneiwitten, het cytoskelet architectuur en moleculaire motoren 1, 2. Bijvoorbeeld, borstklier epitheelcellen (MEC) vormen normale acinaire parenchym wanneer gekweekt op 150 Pa substraten die vergelijkbaar is met de stijfheidgezonde borstklierweefsel. Interessant is dat ze de kenmerken van een zich ontwikkelende tumor, zowel structureel als transcriptie, indien gekweekt op stijver substraten (> 5000 Pa), die de stijfheid van een tumor stroma 8 nabootsen. Bovendien, een ander experiment toont aan dat borst tumorigenese gepaard gaat met collageen verknoping en ECM verstijving 9. Recente experimenten tonen dat humane colon carcinoma (HCT-8) cellen vertonen metastase fenotype (MLP) wanneer zij gekweekt op 2D substraten met fysiologisch relevante stijfheid (20-47 kPa), maar niet op zeer stijf (3,6 GPa) substraten 10- 12 .Deze cellen eerste vorm tumorachtige celclusters en dissociëren van elkaar, vanaf de periferie. Aangezien dit epitheliale naar afgeronde morfologische (E R overgang) verandering optreedt, ze vermenigvuldigen, te verminderen cel-cel en cel-ECM adhesie, en word trekvogels. HCT-8 cellen gekweekt op heel hard polystyreen substraten zijn deze niet vertonenkwaadaardige trekken. Dus het is de hypothese dat HCT-8 cellen uitgezaaide vanwege hun blootstelling aan geschikte micro-omgeving. Vermeldenswaard is dat deze experimenten uitgevoerd met geïmmortaliseerde kankercellijnen of muizen afgeleide primaire cellen, niet met primaire humane kankercellen worden uitgevoerd.
Een recent onderzoek stelt aangevulde cellulaire traction stress, kunnen worden toegepast als een mogelijke biofysische signatuur voor metastatische cellen 13 .De studie omvat het meten trekkracht voor verschillende humane kankercellijnen op polyacrylamide gels. Het blijkt dat metastatische kankercellen significant hoger traction stress kan uitoefenen in vergelijking met niet-metastatische cellen in alle gevallen 13. Echter, deze resultaten direct in tegenspraak met de eerder gepubliceerde bevindingen muizen afgeleide borstkankercellijnen 14. Ook een recent onderzoek ook opvallende verschillen tussen geïmmortaliseerde en primaire humane cellen in het cytoskelet remodeling proeiwit profilering en overleving van de cel eiwit expressie 15. Daarom is het belangrijk voor veel van de biofysische analyses inclusief tractie voor primaire humane kankercellen opnieuw. Dit zal de vraag te beantwoorden of de primaire cellen recapituleren geïmmortaliseerde kanker cellijnen tractie trend.
De hier beschreven protocol geoptimaliseerd voor isolatie van primaire humane colon cellen (zowel gezonde en kanker), en ze te kweken op zachte substraten (polyacrylamide hydrogels) en op petrischalen. Het protocol is gebaseerd op de spijsvertering en de daaruit voortvloeiende enzymatische dissociatie van chirurgische weefselmonster in enkele celsuspensie 16. Voor zover wij weten is dit de eerste demonstratie van het kweken geïsoleerde primaire colon tumor en normale cellen direct op zachte hydrogel substraten met ingebedde fluorescente microkraaltjes tractie cytometrie. Transparante gel substraten immunokleuring ook mogelijk. Deze test toonde verschillen in F-actine organisatiefocale adhesies in primaire menselijke dikke darm cellen als substraat stijfheid veranderingen. Dit celcultuur platform opent de mogelijkheid van het verkennen van de verschillende biofysische eigenschappen van primaire menselijke cellen zoals mobiele stijfheid en tractie als parameters voor kanker prognostiek.
Cellular tractie spanning is onlangs naar voren gekomen als een potentiële biofysische indicator van metastatische staat 13. Echter, geen experimentele tractie gegevens met primaire tumorcellen bestaat in de literatuur tot nu toe. Ook wordt direct kweken geïsoleerde primaire colon cellen op verschillende stijfheid polyacrylamidegelen nog niet gemeld.
