Dupuytrens sykdom (DD) er en fibroproliferative sykdom i håndflaten. Her presenterer vi en protokoll til kultur reseksjonsnivå prøver fra DD i en tredimensjonal (3D) kultur system. Slike korttidskultur systemet tillater preservering av 3D-strukturen og molekylære egenskaper av fibrotisk vev.
Organ fibrosis or “scarring” is known to account for a high death toll due to the extensive amount of disorders and organs affected (from cirrhosis to cardiovascular diseases). There is no effective treatment and the in vitro tools available do not mimic the in vivo situation rendering the progress of the out of control wound healing process still enigmatic.
To date, 2D and 3D cultures of fibroblasts derived from DD patients are the main experimental models available. Primary cell cultures have many limitations; the fibroblasts derived from DD are altered by the culture conditions, lack cellular context and interactions, which are crucial for the development of fibrosis and weakly represent the derived tissue. Real-time PCR analysis of fibroblasts derived from control and DD samples show that little difference is detectable. 3D cultures of fibroblasts include addition of extracellular matrix that alters the native conditions of these cells.
As a way to characterize the fibrotic, proliferative properties of these resection specimens we have developed a 3D culture system, using intact human resections of the nodule part of the cord. The system is based on transwell plates with an attached nitrocellulose membrane that allows contact of the tissue with the medium but not with the plastic, thus, preventing the alteration of the tissue. No collagen gel or other extracellular matrix protein substrate is required. The tissue resection specimens maintain their viability and proliferative properties for 7 days. This is the first “organ” culture system that allows human resection specimens from DD patients to be grown ex vivo and functionally tested, recapitulating the in vivo situation.
Dupuytrens sykdom (DD), en godartet fibroproliferative sykdommen forårsaker permanent strekking av fingrene på grunn av dannelse av knuter og snorer i håndflaten. Selv om sykdommen spredningen er særlig høy blant kaukasiere i Nord-Europa, den underliggende genetisk etiologi av sykdommen er fortsatt ukjent en. Våre siste arbeidet med fibrose har ført oss til utviklingen av enny 3D-kultur-system som gjør det mulig kortvarig kultur av human fibrotisk vev med potensialet av legemiddeltesting. Dette systemet har bidratt til å overvinne den begrensende tilnærming av 2D fibroblastkulturer og å definere en rolle for den delvise nedregulering, oppnås ved ekson hoppetau for aktivering TGFp banen i å mediere fibrose 5.
Vi har utviklet en metode for å kultur ex vivo human reseksjon prøver fra DD-pasienter for å studere interaksjonen mellom myofibroblasts og den omgivende ECM 5, 6. Studiet av DD bindevev fibrose, så vel som andre fibrotiske sykdommer som er avhengig av histopatologisk analyse av det utskårede kirurgisk prøver, isolering av fibroblaster fra vevet og etablering av primære cellekulturer eller sorteringsprosedyrer. Disse metodene er ganske statisk, siden de ikke tillater eksogene manipulasjon av sykdoms egenskaper eller terapeutisk intervensjon ved eksperimentator. I tillegg primær cell kulturer har en tendens til å tilpasse seg forholdene kultur og deres genuttrykk egenskaper avvike vesentlig fra in vivo situasjon ved hver passering, selv under tidlige passasjer (blant passasje 3 og 6) 7, 8. Vi har klart å opprettholde avfall kirurgisk materiale i ex vivo dyrkningsbetingelser for en tidsperiode som tillater undersøkelse av pasientspesifikke egenskaper og screening av anti-fibrotiske eller anti-inflammatoriske medikament-forbindelser.
Systemet er basert på en nitrocellulosemembran som tillater kontakt av vevet med mediet, men ikke med plast, og dermed hindre endring av vev ved festing, som tidligere observert ved dyrkning DD fibroblaster, så vel som andre celletyper 9. Ingen kollagen-gel eller andre ECM-protein-substrat er nødvendig, siden DD vevet i seg selv produserer store mengder av disse proteiner. Dette er fordelaktig for opprettholdelse av nativt ECM mikromiljøet og omsetning syndce matrikssubstrater er viktig regulator av vev arkitektur og funksjon 10, 11. For eksempel ECM-proteiner som fibronektin, laminin og kollagen, kan påvirke front-bak polaritet av fibroblaster som på samme måte som vises for apikal-basal polaritet i epitelceller 12, 13 . Polarisert celler har asymmetrisk fordeling av ekstracellulære molekyler som bestemmer cellemigrasjon og genuttrykk, f.eks α1β1 inte tilgjengelighet på membranen påvirker celleadhesjonen å skrive kollagen 14. Siden en primære målet med denne 3D-modellen var å bevare den opprinnelige mikromiljøet, ble ingen kunstige ECM matrise underlaget brukes.
I korte trekk: reseksjon prøvene er likt delt i et sterilt miljø og plassert på nitrocellular membraner. Ved behandling administreres via injeksjon er nødvendig vevene blir injisert etter at de har blitt plassert på membranen. Hvis behandlingen ikke krever å bli administrert via injection deretter forbindelsen blir tilsatt til kulturmediet (Dulbeccos modifiserte Eagles medium (DMEM), med 1% føtalt kalveserum (FCS), 1% penicillin-streptomycin (P / S)). Kulturene ble opprettholdt i maksimalt ti dager, hvoretter vevet er fiksert i 4% paraformaldehyd (PFA), behandlet gjennom 30% sukrose-oppløsning, innebygd i oktober forbindelse og lagret ved -80 ° C, som tidligere beskrevet 5.
