The fenestrated liver sinusoidal endothelial cell is a biologically important filter system that is highly influenced by various diseases, toxins, and physiological states. These changes significantly impact on liver function. We describe methods for the standardisation of the measurement of the size and number of fenestrations in these cells.
Liver sinusoidal endothelial cells are the gateway to the liver, their transcellular fenestrations allow the unimpeded transfer of small and dissolved substances from the blood into the liver parenchyma for metabolism and processing. Fenestrations are dynamic structures – both their size and/or number can be altered in response to various physiological states, drugs, and disease, making them an important target for modulation. An understanding of how LSEC morphology is influenced by various disease, toxic, and physiological states and how these changes impact on liver function requires accurate measurement of the size and number of fenestrations. In this paper, we describe scanning electron microscopy fixation and processing techniques used in our laboratory to ensure reproducible specimen preparation and accurate interpretation. The methods include perfusion fixation, secondary fixation and dehydration, preparation for the scanning electron microscope and analysis. Finally, we provide a step by step method for standardized image analysis which will benefit all researchers in the field.
As células endoteliais sinusoidais hepáticas (LSECs) são altamente diferenciadas, células endoteliais que revestem a parede da sinusóide hepática. LSECs são perfuradas com fenestrações que são não-diaphragmed, transcelular poros de 50-250 nm de diâmetro. Até 20% da superfície é coberta por LSECs fenestrações, que são geralmente em grupos de dezenas a centenas chamadas placas de peneira 1-3 (Figura 1). O estudo de fenestrações é difícil. O diâmetro das fenestrações se encontra abaixo da resolução da microscopia óptica convencional, de modo que anteriormente apenas a observação por microscopia de electrões, tanto em tecido do fígado ou cultivadas LSECs intactos tem sido possível. O microscópio eletrônico de varredura (SEM) tem sido mais frequentemente utilizado para estudar tamanho fenestration, a frequência ea porosidade (a percentagem de membrana LSEC que é perfurado por fenestrations) porque SEM permite a observação de grandes áreas de superfície endotelial e medição de milhares, se não dezenas de milhares de fenestrações. Apesar de sua utilidade, os resultados que são relatados a partir de estudos baseados SEM-paraLSEC parâmetros como o tamanho fenestra�o, número, frequência e a porosidade pode variar amplamente na literatura (Quadro 1).
Fenestrações e pratos perfurados são estruturas frágeis que contrato, quebram, dilatam ou se aglutinam durante a preparação de amostras, processamento, assim, o cuidado é necessário para preservar a sua integridade. A pressão de perfusão elevada 14; osmolaridade incorreto do fixador e buffers 15; fixação inadequada ou fixação do tempo; e velocidade de desidratação pós-fixação e secagem são todas as áreas de processamento para SEM que podem produzir produtos manufacturados que interferem com a preservação da ultraestrutura (Figura 2). Perda de fenestrações ('defenestration') e fenestra�o encolhimento pode ocorrer como resultado de má fixação, resultando na redução de diâmetro e porosidade fenestra�o célula. Métodos para melhorar a conservação de amostras para análise em MEV foram descritos anteriormente 15-17 e será discutidoaqui com dicas adicionais sobre como melhorar a preservação da amostra. Os principais objectivos da preservação espécime são para remover o sangue dos sinusóides, de modo que a superfície do LSEC pode ser visualizado e para evitar danos ou LSEC de alta pressão ou fixação retardada. Perfusão do fígado inteiro de fixador através da veia portal é o método preferido para a fixação hepática. Tal como descrito em pormenor noutro 16,18 perfusão deve ser efectuada a baixa pressão (por exemplo, 10 cm de H 2 0) para evitar artefactos relacionados com a pressão de perfusão e os danos para a LSEC, tipicamente manifestada como grandes lacunas dentro da membrana celular. No entanto, muitas vezes fixação razoável pode ser obtido utilizando a agulha de perfusão de biópsias de fígado de seres humanos e animais, tal como descrito em detalhe noutro local 19. Nós também apresentamos um método de análise de imagem que minimiza a inclusão de artefactos, e padroniza a medição das fenestrações. Variação na seleção de senóides para micrografias, análise de imagens de artefatos, e medição de área celular para porosidade e fenestration frequência levaram a grandes discrepâncias nos resultados publicados. A abordagem padronizada de avaliação e medida de fenestrações e os requisitos mínimos para a apresentação dos dados não tenham sido claramente abordada na literatura anteriormente 4,10,20-31.
A capacidade de medir de forma precisa e reprodutivelmente o estado do endotélio sinusoidal fígado é um passo importante na compreensão da biologia destas células altamente especializadas.
