The fenestrated liver sinusoidal endothelial cell is a biologically important filter system that is highly influenced by various diseases, toxins, and physiological states. These changes significantly impact on liver function. We describe methods for the standardisation of the measurement of the size and number of fenestrations in these cells.
Liver sinusoidal endothelial cells are the gateway to the liver, their transcellular fenestrations allow the unimpeded transfer of small and dissolved substances from the blood into the liver parenchyma for metabolism and processing. Fenestrations are dynamic structures – both their size and/or number can be altered in response to various physiological states, drugs, and disease, making them an important target for modulation. An understanding of how LSEC morphology is influenced by various disease, toxic, and physiological states and how these changes impact on liver function requires accurate measurement of the size and number of fenestrations. In this paper, we describe scanning electron microscopy fixation and processing techniques used in our laboratory to ensure reproducible specimen preparation and accurate interpretation. The methods include perfusion fixation, secondary fixation and dehydration, preparation for the scanning electron microscope and analysis. Finally, we provide a step by step method for standardized image analysis which will benefit all researchers in the field.
De lever sinus endotelceller (LSECs) är mycket differentierade endotelceller som kantar väggen av leversinus. LSECs är perforerade med fenestrationer som är icke-diaphragmed, porer transcellulär 50-250 nm i diameter. Upp till 20% av ytan på LSECs är täckt av fenestrationer, som vanligtvis i grupper om tiotals till hundratals kallade sållningsplattor 1-3 (Figur 1). Fenestrationer möjliggöra överföring av plasma och nanosubstrates mellan blod och hepatocyter, vilket skapar en mycket effektiv ultrafiltreringssystem. Fenestrationer är dynamiska strukturer – både deras storlek och / eller nummer kan ändras som svar på olika fysiologiska tillstånd, läkemedel och sjukdomar. Till exempel, fenestrationer är större i den fastande än i födointag 4; 2-di-iodoamphetamine ökar fönsternummer, 5,6 och en minskning i storlek och antal fenestrationer per cell sker i åldrande och många sjukdomstillstånd 7-13 </sup>. Noggrann mätning av storleken och antalet fenestrationer är viktigt för att förstå hur LSEC morfologi påverkas av olika sjukdomar, giftiga, och fysiologiska tillstånd; deras inverkan på leverfunktionen; och för att utveckla fenestration modulerande terapeutiska insatser 1.
Studien av fenestrationer är svårt. Diametern på fenestrationer ligger under upplösningen av konventionella ljusmikroskop, så tidigare bara observation med hjälp av elektronmikroskopi både i intakta levervävnad eller odlade LSECs har varit möjligt. Den svepelektronmikroskop (SEM) har oftast använts för att studera fönsterstorlek, frekvens och porositet (andelen LSEC membran som är perforerad av fenestrationer) eftersom SEM möjliggör observation av stora delar av endotelytan och mätning av tusentals, om inte tiotusentals fenestrationer. Trots dess användbarhet, de resultat som rapporterats från SEM baserade studier förLSEC parametrar såsom fönsterstorlek, antal, frekvens och porositet varierar kraftigt i litteraturen (Tabell 1).
Fenestrationer och silplattor är ömtåliga strukturer som kontraktet bryts, vidgar eller smälter samman under extraktion behövs därför noggrann behandling för att bevara sin integritet. Förhöjda perfusionstryck 14; felaktig osmolaritet fixativ och buffertar 15; otillräcklig fixering eller fixering tid; och hastighet efter fixering dehydratisering och torkning är alla områden av behandling för SEM som kan producera artefakter som stör bevarandet av ultrastrukturen (Figur 2). Förlust av fenestrationer (defenestration ") och fönster krympning kan uppstå som en följd av dålig fixering, vilket resulterar i minskad fenestration diameter och cell porositet. Metoder för att förbättra bevarandet av exemplar för SEM-analys har beskrivits tidigare 15-17 och kommer att diskuterashär med ytterligare tips om hur man kan förbättra prov bevarande. De viktigaste målen för provet bevarande är att ta bort blod från sinusoids så att ytan av LSEC kan visualiseras och för att undvika LSEC skador från antingen högt tryck eller fördröjd fixering. Hela leverperfusion fixativ via portalen ven är den lämpligaste metoden för lever fixering. Såsom beskrivs i detalj på annat ställe 16,18 perfusionen måste genomföras vid lågt tryck (t ex 10 cm av H 2 0) för att undvika tryckrelaterade perfusion artefakter och skador på LSEC, typiskt visar sig som stora luckor inom i cellmembranet. Emellertid kan rimlig fixering ofta erhållas med användning av nål perfusion av leverbiopsier från människor och djur, såsom beskrivs i detalj på annan plats 19. Denna teknik innebär att direkt injicera fixativ i vävnaden tills blodet spolas ut ur provet och vävnaden är fast och fast. Fixering av prover för elektronmikroskopi måste vara perfOrmed så snabbt som möjligt efter avslutad blodflödet för att förhindra ultra förändringar som sker som en följd av de lever extremt snabba autolytiska processer.
Vi presenterar också en metod för bildanalys som minimerar införandet av artefakter, och standardiserar mätningen av fenestrationer. Variation i valet av sinus för mikrofotografier, bildanalys av artefakter, och mätning av cellområdet för porositet och fönsterfrekvens har lett till stora skillnader i publicerade resultat. En standardiserad metod för utvärdering och mätning av fenestrationer och de minimikrav för datapresentation har inte klart upp i litteraturen tidigare 4,10,20-31.
Förmågan att noggrant och reproducerbart mäta status levern sinus endotel är ett viktigt steg i att förstå biologi dessa mycket specialiserade celler. Nyare tekniker såsom strukturerad belysning mikroskopi 32, atomkraftsmikroskopi 33 och d-STORM (direkt Stochastic Optical Reconstrucion Microscopy) 34 kommer att ge viktig information om morfologin hos dessa celler in vitro men SEM fortfarande den främsta metoden för att visualisera och mäta deras struktur i situ.</e…
