Optically transparent zebrafish embryos are widely used to study and visualize in real time the interactions between pathogenic microorganisms and the innate immune cells. Micro-injection of Mycobacterium abscessus, combined with fluorescence imaging, is used to scrutinize essential pathogenic features such as cord formation in zebrafish embryos.
Zebrafish (Danio rerio) embryos are increasingly used as an infection model to study the function of the vertebrate innate immune system in host-pathogen interactions. The ease of obtaining large numbers of embryos, their accessibility due to external development, their optical transparency as well as the availability of a wide panoply of genetic/immunological tools and transgenic reporter line collections, contribute to the versatility of this model. In this respect, the present manuscript describes the use of zebrafish as an in vivo model system to investigate the chronology of Mycobacterium abscessus infection. This human pathogen can exist either as smooth (S) or rough (R) variants, depending on cell wall composition, and their respective virulence can be imaged and compared in zebrafish embryos and larvae. Micro-injection of either S or R fluorescent variants directly in the blood circulation via the caudal vein, leads to chronic or acute/lethal infections, respectively. This biological system allows high resolution visualization and analysis of the role of mycobacterial cording in promoting abscess formation. In addition, the use of fluorescent bacteria along with transgenic zebrafish lines harbouring fluorescent macrophages produces a unique opportunity for multi-color imaging of the host-pathogen interactions. This article describes detailed protocols for the preparation of homogenous M. abscessus inoculum and for intravenous injection of zebrafish embryos for subsequent fluorescence imaging of the interaction with macrophages. These techniques open the avenue to future investigations involving mutants defective in cord formation and are dedicated to understand how this impacts on M. abscessus pathogenicity in a whole vertebrate.
Mycobacterium abscessus er en ny patogen som forårsaker et vidt spektrum av kliniske syndromer hos mennesker. Disse inkluderer kutane infeksjoner samt alvorlige kroniske lungeinfeksjoner, for det meste møtt i immunsvikt og pasienter med cystisk fibrose 1,2,3,4. M. abscessus regnes også som en stor raskt voksende mykobakterielle arter som er ansvarlige for nosokomiale og iatrogenic infeksjoner hos mennesker. Dessuten flere siste rapportene fremhevet muligheten for at M. abscessus kan krysse blod-hjerne-barrieren og induserer viktige lesjoner i sentralnervesystemet (CNS) 5,6. Til tross for at en rask dyrker, M. abscessus utstillinger også flere sykdomsfremkallende funksjoner som er relatert til de av Mycobacterium tuberculosis, inkludert evnen til å tie for år innenfor granulomatøs strukturer og å generere caseous lesjoner i lungene 7. Mer alarmerende er den lave sensensitivitet på M. abscessus mot antibiotika, gjengi disse infeksjonene ekstremt vanskelige å behandle fører til en betydelig terapeutisk strykprosent 8,9. Den viktigste trusselen av denne arten er i hovedsak sin iboende motstand mot antibiotika, som er av stor bekymring i offentlige helseinstitusjoner 10 og en kontraindikasjon for lungetransplantasjon 11.
M. abscessus skjermer glatte (S) eller grov (R) koloni morfotyper som fører til forskjellige kliniske utfall. I motsetning til den S-stammen, R bakterier har en tendens til å vokse ende til ende, noe som fører til et tau eller snor-lignende struktur 12,13. Flere uavhengige studier basert på enten mobil eller dyremodeller avslørte hyper-virulens fenotype av R morphotype 14,15. Fra epidemiologiske studier, de mest alvorlige tilfeller av M. abscessus lungeinfeksjoner synes å være assosiert med R-variantene 16 som er den eneste varianten somhar blitt sett å vedvare i mange år i en infisert vert tre. Den morphotype Forskjellen er avhengig av tilstedeværelse (i S) eller tap (i R) av overflate forbundet glycopeptidolipids (GPL) 12. Men på grunn av de iboende begrensninger av de tilgjengelige cellulære / dyremodeller som brukes til å studere M. abscessus infeksjon, vår kunnskap om patofysiologiske hendelsene i de R eller S varianter er fortsatt uklar. Infeksjon av immuno-kompetente mus via intravenøse eller spray ruter fører til forbigående kolonisering, som hindrer bruk av mus for å studere vedvarende infeksjoner og for in vivo resistenstesting 17. Derfor er utviklingen av dyremodeller mottagelig for manipulering av vertsrespons er en stor utfordring. I denne sammenheng har ikke-pattedyr-modeller av infeksjon nylig utviklet, herunder Drosophila melanogaster 18 som har flere fordeler, slik som pris, hastighet og etisk aksept over musemodell. Sebrafisk (Danio rerio) modell for infeksjon har også blitt undersøkt for å visualisere, av ikke-invasiv bildediagnostikk, progresjon og kronologi av M. abscessus infeksjon i et levende dyr 19. Viktigere, et bevis på konseptet ble også etablert for å demonstrere sin egnethet for in vivo antibiotika vurderingene mot M. abscessus 17,20.
Sebrafisk har blitt mye brukt i løpet av de siste to tiårene for å studere samspillet mellom ulike patogener og vertens immunsystemet 21. Den økende suksess med dette alternativet virveldyr modellen baserer seg på store og unike muligheter som motiverte og godkjente bruken for en bedre forståelse av en rekke virus og bakterieinfeksjoner 19,22,23,24,25,26,27,28,29. I motsetning til de fleste andre dyremodeller, sebrafisk embryo er optisk transparent, slik at ikke-invasiv fluorescens bildebehandling 30. Dette HAs ledet til å studere M. abscessus smittet sebrafisk embryo med enestående detaljer, som kulminerte med beskrivelsen av ekstracellulære snor, som representerer et eksempel på bakteriell morfologisk plastisitet. Kordeller representerer en ny mekanisme for undergraving av immunsystemet, og en nøkkelmekanisme fremme patogenese av akutt M. abscessus infeksjon 19.
Denne rapporten beskriver nye verktøy og metoder ved hjelp av sebrafisk embryo å tyde patofysiologiske trekk av M. abscessus smitte og å studere de intime samspillet mellom basiller og det medfødte immunsystemet. Først en detaljert mikroinjeksjon protokoll som omfatter behandling av bakteriepodestoff, embryo forberedelse, og infeksjoner per se, er presentert. Metoder spesielt tilpasset for å vurdere M. abscessus virulens ved å måle ulike parametre, som vert overlevelse og bakteriemengden, blir presentert. Spesiell fokus er gitt på hvordanfor å overvåke, ved et nivå i tid og rom, skjebnen og progresjon av infeksjonen og vertens immunrespons mot M. abscessus bruker videomikroskopi. Videre å undersøke bidrag og rolle makrofager under M. abscessus infeksjon, metoder generere makrofager-utarmet embryoer (ved hjelp av enten genetically- eller kjemisk-baserte tilnærminger) er beskrevet. Til slutt, for å protokoller visualisere de spesifikke interaksjoner med makrofager eller nøytrofile bruker enten fast eller levende embryoer er dokumentert.
Målet med denne rapporten er å stimulere videre studier for å kaste nytt lys inn M. abscessus virulensmekanismer og særlig rolle snor i etableringen av en akutt infeksjon og ukontrollert prosess.
Sebrafisk har nylig dukket opp som en utmerket virveldyr modellsystem for å studere dynamikken i bakterieinfeksjon ved hjelp av bredt felt og confocal bildebehandling i sanntid 36. Kombinasjonen av dispergert mykobakterielle suspensjoner (protokoll 2.2) sammen med mikroinjeksjonsmetoder (protokoll 4) tillater reproduserbare systemiske infeksjoner, og etterfølgende overvåking og visualisering av progresjon av infeksjon med et spesielt fokus på de bakterielle interaksjoner med vertsmakrofager.
