A method for imaging changes in membrane potential using genetically encoded voltage indicators is described.
Genetically encoded voltage indicators (GEVIs) have improved to the point where they are beginning to be useful for in vivo recordings. While the ultimate goal is to image neuronal activity in vivo, one must be able to image activity of a single cell to ensure successful in vivo preparations. This procedure will describe how to image membrane potential in a single cell to provide a foundation to eventually image in vivo. Here we describe methods for imaging GEVIs consisting of a voltage-sensing domain fused to either a single fluorescent protein (FP) or two fluorescent proteins capable of Förster resonance energy transfer (FRET) in vitro. Using an image splitter enables the projection of images created by two different wavelengths onto the same charge-coupled device (CCD) camera simultaneously. The image splitter positions a second filter cube in the light path. This second filter cube consists of a dichroic and two emission filters to separate the donor and acceptor fluorescent wavelengths depending on the FPs of the GEVI. This setup enables the simultaneous recording of both the acceptor and donor fluorescent partners while the membrane potential is manipulated via whole cell patch clamp configuration. When using a GEVI consisting of a single FP, the second filter cube can be removed allowing the mirrors in the image splitter to project a single image onto the CCD camera.
L'objectif principal de cet article est de démontrer l'imagerie optique de l'évolution des potentiels de membrane in vitro utilisant des protéines fluorescentes codées génétiquement. changements d'imagerie du potentiel de membrane offre la possibilité intéressante de l'étude de l'activité des circuits neuronaux. Lorsque des changements dans la membrane résultat potentiel à un changement d'intensité de fluorescence, chaque pixel de la caméra devient une électrode de substitution permettant des mesures non intrusives de l'activité neuronale. Depuis plus de quarante ans, des colorants sensibles à la tension organiques ont été utiles pour observer les changements de potentiel 1-4 membrane. Cependant, ces colorants manquent de spécificité cellulaire. En outre, certains types de cellules sont difficiles à teindre. indicateurs de tension génétiquement codés (GeViS) à surmonter ces limitations en ayant les cellules à étudier spécifiquement expriment la sonde fluorescente sensible à la tension.
Il existe trois classes de GeViS. La première classe de GEVI utilise la vodomaine de la phosphatase de détection de tension nsion détection soit avec une seule protéine fluorescente (FP) 5-9 ou un transfert d'énergie par résonance Förster (FRET) paire 10-12. La seconde catégorie de capteurs utilise microbienne rhodopsine comme un indicateur fluorescent directement ou par l'intermédiaire de 13 à 15 FRET électrochrome 16,17. La troisième classe utilise deux composantes, la composante génétique étant une membrane ancrée FP et un second composant étant une membrane lié trempe colorant 18-20. Tandis que les deuxième et troisième catégories sont utiles pour in vitro et trancher les expériences 19,20, seule la première classe de capteurs sont actuellement utile pour des analyses in vivo 6.
Dans ce rapport, nous allons démontrer l'imagerie du potentiel de membrane en utilisant la première classe de GeViS (Figure 1) in vitro. Cette première classe de capteurs de tension est le plus facile de faire la transition à l'imagerie in vivo. Depuis GeViS utilizing un domaine de détection de tension fusionné à un FP sont environ 50 fois plus brillante que la classe de rhodopsine de capteurs, ils peuvent être imagés en utilisant un éclairage à lampe à arc au lieu d'exiger un laser extrêmement puissant. Une autre conséquence de l'écart de brillance qui est la première classe de GeViS peut facilement dépasser l'auto-fluorescence du cerveau. Les sondes à base de rhodopsine ne peut pas. La troisième classe de capteur est tout aussi brillante que la première catégorie, mais nécessite l'ajout d'un extincteur chimique qui est difficile à administrer in vivo.
Nous allons donc démontrer l'acquisition d'une sonde avec une seule FP (Bongwoori) 8 et une sonde constituée d'une paire de FRET (Nabi 2) 12. Le FRET construit dans le présent rapport sont des versions de papillon de VSFP-CR (protéines fluorescentes sensibles à la tension – Clover-mRuby2) 11 consistant en un donneur fluorescente verte, Clover, et un accepteur de fluorescence rouge, mRuby2, nommé Nabi 2.242 et 2.244 Nabi <sjusqu'à> 12. L'introduction de ces types d'enregistrements devrait permettre aux chercheurs de mieux comprendre le type de GeViS d'information peut fournir.

Le système nerveux utilise une tension de plusieurs manières différentes, l'inhibition entraîne une légère hyperpolarisation, entrée synaptique provoque une légère dépolarisation et une action résultats potentiels d'une variation relativement importante de tension.
