Eliminating specific cells without damaging other cells is extremely difficult, especially in established tissue, yet there is an urgent need for a cell elimination method in the tissue engineering field. Here, we present a method for specific cell elimination from a mixed 3D cell culture using near infrared photoimmunotherapy (NIR-PIT).
Recent developments in tissue engineering offer innovative solutions for many diseases. For example, tissue engineering using induced pluripotent stem cell (iPS) emerged as a new method in regenerative medicine. Although this tissue regeneration is promising, contamination with unwanted cells during tissue cultures is a major concern. Moreover, there is a safety concern regarding tumorigenicity after transplantation. Therefore, there is an urgent need for eliminating specific cells without damaging other cells that need to be protected, especially in established tissue. Here, we present a method for specific cell elimination from a mixed 3D cell culture in vitro with near infrared photoimmunotherapy (NIR-PIT) without damaging non-targeted cells. This technique enables the elimination of specific cells from mixed cell cultures or tissues.
A eliminação de células específicas, sem danificar outras células é extremamente difícil, especialmente no tecido estabelecido, e não há uma necessidade urgente de um método de eliminação de células no campo da engenharia de tecidos. Hoje em dia no campo da medicina regenerativa, as culturas de tecido, utilizando células estaminais embrionárias (ES), células estaminais pluripotentes (PSCs), ou induzida por células estaminais pluripotentes (IPS) são materiais promissores 1-3.
Embora esta regeneração de tecidos é promissor, a contaminação com células não desejadas é uma grande preocupação. Além disso, existe uma preocupação de segurança de tumorigenicidade após o transplante 4,5. Embora muitos estudos têm-se centrado sobre estas questões para eliminar as células específicas, especialmente em medicina regenerativa 6-8, nenhum método prático tem sido desenvolvido.
Perto photoimmunotherapy infravermelho (NIR-PIT) é um tratamento baseado em um anticorpo conjugat-fotoabsorventee (APC). Uma APC é constituído por um anticorpo específico de células monoclonais (mAb) e um fotoabsorvente, IR700. IR700 é um derivado de ftalocianina de sílica-hidrofílico e não induz por si só a fototoxicidade 9. IR700 é covalentemente conjugada com o anticorpo através de resíduos de amida na cadeia lateral de moléculas de lisina. A APC liga-se moléculas alvo na membrana celular e, em seguida, induz necrose celular quase imediato após a exposição à luz NIR a 690 nm. Durante a exposição à NIR-luz, as rupturas de membranas celulares que levam à morte celular 9-14. NIR-PIT tem provado ser eficazes com vários anticorpos ou fragmentos de anticorpos, incluindo o anti-EGFR, anti-HER2, anti-PSMA, anti-CD25, anti-mesotelina, anti-GPC3, e anti-CEA 15-21. Portanto, NIR-poço pode ser usada contra uma ampla variedade de moléculas-alvo. Além disso, NIR-PIT é um tratamento bem controlado que permite tratamento selectivo de regiões específicas, restringindo o NIR-light irradiação 18,22.

Nós demonstramos um método de eliminação de células específicas a partir de uma cultura de células mista 3D sem danos para as células não alvo usando NIR-poço.
