Eliminating specific cells without damaging other cells is extremely difficult, especially in established tissue, yet there is an urgent need for a cell elimination method in the tissue engineering field. Here, we present a method for specific cell elimination from a mixed 3D cell culture using near infrared photoimmunotherapy (NIR-PIT).
Recent developments in tissue engineering offer innovative solutions for many diseases. For example, tissue engineering using induced pluripotent stem cell (iPS) emerged as a new method in regenerative medicine. Although this tissue regeneration is promising, contamination with unwanted cells during tissue cultures is a major concern. Moreover, there is a safety concern regarding tumorigenicity after transplantation. Therefore, there is an urgent need for eliminating specific cells without damaging other cells that need to be protected, especially in established tissue. Here, we present a method for specific cell elimination from a mixed 3D cell culture in vitro with near infrared photoimmunotherapy (NIR-PIT) without damaging non-targeted cells. This technique enables the elimination of specific cells from mixed cell cultures or tissues.
Eliminando cellule specifiche senza danneggiare altre cellule è estremamente difficile, soprattutto nel tessuto stabilita, e vi è una necessità urgente di un metodo di eliminazione di cella nel campo dell'ingegneria dei tessuti. Al giorno d'oggi nel campo della medicina rigenerativa, colture di tessuti utilizzando cellule staminali embrionali (ES), le cellule staminali pluripotenti (PSC), o cellule staminali pluripotenti indotte (iPS) sono materiali promettenti 1-3.
Anche se questa rigenerazione dei tessuti è promettente, la contaminazione con cellule indesiderate è una preoccupazione importante. Inoltre, vi è un problema di sicurezza di tumorigenicità dopo trapianto 4,5. Anche se molti studi si sono concentrati su questi temi per eliminare cellule specifiche, in particolare nel campo della medicina rigenerativa 6-8, nessun metodo pratico è stato sviluppato.
Vicino photoimmunotherapy infrarosso (NIR-PIT) è un trattamento a base di un anticorpo conjugat-photoabsorbere (APC). Un APC è costituito da un anticorpo specifico per cellule monoclonali (mAb) e un photoabsorber, IR700. IR700 è un derivato del idrofila di silice-ftalocianina e non induce di per sé fototossicità 9. IR700 è covalentemente coniugato all'anticorpo tramite residui ammidici sulla catena laterale di molecole di lisina. L'APC si lega molecole bersaglio sulla membrana cellulare e quindi induce necrosi cellulare quasi immediatamente dopo l'esposizione alla luce NIR a 690 nm. Durante l'esposizione a NIR luce, la rottura della membrana cellulare che portano alla morte batteria 9 – 14 anni. NIR-PIT ha dimostrato di essere efficace con più anticorpi o frammenti di anticorpi, tra cui anti-EGFR, anti-HER2, anti-PSMA, anti-CD25, anti-mesotelina, anti-GPC3, e anti-CEA 15 – 21. Pertanto, NIR-PIT può essere utilizzato contro un'ampia varietà di molecole bersaglio.
