We present a biochemical and behavioral protocol to evaluate the efficacy of mitochondria-targeted water-soluble compounds for the treatment of Spinocerebellar ataxia type 1 (SCA1) and other cerebellar neurodegenerative diseases.
Mitochondriale dysfunctie speelt een belangrijke rol bij het verouderingsproces en neurodegeneratieve ziekten waaronder verscheidene erfelijke Spinocerebellaire Ataxie en andere bewegingsstoornissen gekenmerkt door progressieve degeneratie van het cerebellum. Het doel van dit protocol is het mitochondriale dysfunctie spinocerebellaire ataxie type 1 (SCA1) evalueren en de effectiviteit van farmacologische doelwit metabole ademhaling evalueren via de in water oplosbare verbinding barnsteenzuur progressie van de ziekte vertragen. Deze benadering geldt voor andere cerebellum ziekten en kan worden aangepast aan een groot aantal in water oplosbare therapieën.
Ex vivo analyse van mitochondriale respiratie wordt gebruikt voor het detecteren en kwantificeren van ziekte gerelateerde veranderingen in mitochondriale functie. Genetische bewijs (ongepubliceerde gegevens) en proteomics bewijs van mitochondriale dysfunctie bij SCA1 muismodel, evalueren we het effect van behandeling met de in water oplosbare metabole booster succinic zuur door het oplossen van deze verbinding direct in de kooi drinkwater. Het vermogen van het geneesmiddel om de bloed-hersenbarrière passeert af te leiden met behulp van hoge prestatie vloeistofchromatografie (HPLC). De werkzaamheid van deze verbindingen kunnen vervolgens worden getest met meerdere gedragsparadigma's waaronder de versnellende rotarod, balk footprint testen en analyse. Cytoarchitectural integriteit van de kleine hersenen kan worden bepaald met behulp van immunofluorescentie assays dat Purkinjecel kernen en Purkinjecel dendrieten en soma te sporen. Deze methoden zijn robuust technieken voor het bepalen van mitochondriale dysfunctie en de werkzaamheid van de behandeling met in water oplosbare verbindingen in cerebellaire neurodegeneratieve ziekte.
Mitochondriën zijn de belangrijkste producenten van adenosine trifosfaat (ATP), een co-enzym essentieel voor cellulaire energie, waarbij het merendeel van mitochondriale ATP geproduceerd door middel van oxidatieve fosforylering (OXPHOS) met de elektronen transportketen. De hersenen, gezien de hoge metabole eisen en de afhankelijkheid van oxidatieve fosforylering voor het voeden van neurale activiteit, is zeer gevoelig voor mitochondriale dysfunctie. Hierdoor wordt mitochondriale dysfunctie geactiveerd tijdens het verouderingsproces 1 en is betrokken bij de pathogenese van verschillende neurodegeneratieve ziekten 2, 3, 4. Hieruit volgt dat mitochondria aantrekkelijke therapeutische doelwitten voor neurodegeneratie.
In dit protocol, hebben wij het gebruik van spinocerebellaire ataxie type 1 (SCA1) als model neurodegeneratieve ziekte vastgesteld voor de studie van mitochondrial dysfunctie en de ontwikkeling van mitochondriale gerichte therapieën. SCA1 wordt veroorzaakt door polyglutamine (polyQ) repeat expansie mutatie in het ataxine-1 genproduct dat de geleidelijke degeneratie van Purkinje neuronen van het cerebellum en neuronen van andere hersengebieden activeert. De transgene muis lijn hier gebruikt (aangeduid als SCA1 muis), die een polyQ-mutant ataxine-1 transgen onder de controle van een Purkinje-celspecifieke promoter tot expressie brengt, maakt de doelgerichte analyse van de Purkinje-cellen component van SCA1 5. SCA1 muizen ondergaan geleidelijke Purkinjecel degeneratie en ontwikkelen ataxic gang 6.
Mitochondriale dysfunctie complex en mitochondriale gerichte behandeling werkzaamheid kan worden geëvalueerd door een batterij van moleculaire en gedrags-assays. Mitochondriële complex dysfunctie wordt gemeten ex vivo door ademhaling testen die veranderde zuurstofverbruik binnen cerebellum weefsel in te detecterende aanwezigheid van elektronen transportketen substraten en remmers 7. Ademhaling assays zijn eerder gebruikt gepermeabiliseerde weefsel, mitochondrial isolaten en gehele weefsel 7, 8, 9. Zij zorgen voor directe beoordeling van de mitochondriale functie in tegenstelling tot morfologische dataverzameling methoden zoals transmissie-elektronenmicroscopie of immunofluorescentiekleuring. Het gebruik van hele weefsels dan geïsoleerde mitochondria voorkomt vooringenomen selectie van gezonde mitochondriën die kunnen optreden tijdens de isolatiewerkwijze 7. Na aanpassing aan het protocol zoals de ademhaling assay is een waardevolle werkwijze voor het detecteren mitochondriale dysfunctie in cerebellaire neurodegeneratieve ziekten.
Niet-specifieke activatoren van metabolisme kan worden gebruikt om mitochondriale dysfunctie afleiden in transgene muizen modellen van neurodegeneratieve disease en hulp bij de ontwikkeling van nieuwe therapieën. Quercetine, co-enzym Q10 en creatine zijn allemaal aangetoond neurodegeneratieve pathologie verbeteren bij patiënten en in diermodellen voor neurodegeneratieve ziekten 10, 11, 12, 13, 14, 15, 16. Hier presenteren we een nieuwe metabole activator, barnsteenzuur, het metabolisme stimuleren en mitochondriale neurodegeneratieve ziekte. Opdat de activator is de bloed-hersenbarrière, HPLC werd toegepast voor levering aan neuraal weefsel te detecteren in 17 behandelde muizen.
Om de therapeutische effecten van metabolisch gerichte water oplosbare verbindingen zoals barnsteenzuur evalueren, kan een batterij van gedragsparadigma's en immunopathologische studies worden gebruikt. due de motorische coördinatie in cerebellaire neurodegeneratieve ziekte, de voetafdruk baan assay beam test en versnellen roterende staaf test worden gebruikt om redding van gedrags- pathologie 6, 18, 19 detecteren. Deze maatregelen worden aangevuld met immunopathologische evaluatie van cerebellaire cytoarchitecture door het beoordelen van moleculaire laagdikte (gedefinieerd als Purkinjecel dendritische prieel lengte) en Purkinjecel soma tellingen binnen een bepaalde kwabje van cerebellaire weefsel 6, 20, 21. Hier presenteren we meerdere neuropathologische en gedragsmatige methoden voor de opsporing en behandeling van mitochondriale dysfunctie met metabolisch gerichte water oplosbare verbindingen.
We maken gebruik van ex vivo analyse van mitochondriale ademhaling tot mitochondriale dysfunctie te analyseren in de SCA1 transgenic muis. Verder laten wij ziekte symptomen en pathologie zijn verbeterd door de in water oplosbare mitochondriale booster barnsteenzuur, verder impliceert mitochondriale dysfunctie in SCA1 ziekteprogressie.
If these methods are used as described, they are capable of detecting and alleviating oxidative phosphorylation-mediated mitochondrial dysfunction in cerebellar neurodegenerative disease mice models. The combined biochemical and behavioral assays are multifaceted methods for determining the extent of mitochondrial contribution to cerebellar neurodegenerative disease pathology.
