The goal of this protocol is to provide users with a set of methods for the high-throughput decapsulation of Lymnaea stagnalis embryos and larvae in preparation for whole mount in situ hybridization, and for subsequent pre- and post-hybridization treatments.

I molluschi sono un gruppo di animali che tengono l'interesse di una vasta diversità di discipline scientifiche. Nonostante la loro diversità morfologica 1, ricchezza di specie (secondo solo a Artropodi in termini di numero di specie 2) e rilevanza per una vasta gamma di commerciali 3, 4 medici e scientifici questioni 5-8, ci sono relativamente poche specie di molluschi che possono vantare di essere entrambi i modelli scientifici ben attrezzate e facili da mantenere in un ambiente di laboratorio. Un mollusco che è molto utilizzato da discipline come la neurobiologia 9, ecotossicologia 10 e la biologia evolutiva, più recentemente 11,12, è Lymnaea stagnalis, soprattutto a causa della sua distribuzione capillare ed estrema facilità di manutenzione. Questa estrazione, o 'decapsulation', deve essere realizzato in modo efficiente per raccogliere materiale adeguato per un dato esperimento in situ, mentre allo stesso tempo mantenere l'integrità morfologica e cellulare. Mentre altri organismi modello subiscono anche lo sviluppo incapsulato, nelle nostre mani nessuno dei metodi riportati per quelle specie potrebbe essere impiegato con successo in L. stagnalis.
Gli obiettivi generali di questo metodo sono quindi: per estrarre L. stagnalis embrioni e larve dalle loro capsule in un modo high-throughput, di applicare i trattamenti pre-ibridazione che ottimizzano il segnale WMISH, per preparare gli embrioni e larve con WMISHsignals soddisfacenti per l'imaging.
Il metodo qui descritto permette la visualizzazione efficiente dei trascritti di RNA con presumibilmente diversi livelli di espressione all'interno di tutte le fasi di sviluppo di Lymnaea stagnalis. Per rimuovere gli embrioni e larve dalle loro capsule abbiamo trialed una varietà di sostanze chimiche, shock osmotico e trattamenti fisici riportati per altri encapsulated- lo sviluppo di organismi modello.
