The goal of this protocol is to provide users with a set of methods for the high-throughput decapsulation of Lymnaea stagnalis embryos and larvae in preparation for whole mount in situ hybridization, and for subsequent pre- and post-hybridization treatments.
Whole mount in situ Hybridisierung (WMISH) ist eine Technik , die für die räumliche Auflösung von Nukleinsäuremolekülen (oft mRNAs) innerhalb eines "whole mount" Gewebepräparation oder Entwicklungsstadium (wie ein Embryo oder Larve) von Interesse ermöglicht. Diese Verfahren umfassen die Zuordnung von Larven unbekannter chronologischen Alter zu einem ontogenetischer Fenster die effiziente Entfernung von Embryonen und Larven von ihren Eier Kapseln, die Anwendung eines geeigneten Proteinase-K – Behandlung für jeden ontogenetischer Fenster und Hybridisierung, post-Hybridisierung und Immunnachweis Schritte. Diese Methoden stellen eine Grundlage, das resultierende Signal für eine bestimmte RNA-Transkript verfeinert werden kann weiter mit Sonde spezifische Anpassungen (in erster Linie Sondenkonzentration und Hybridisierungstemperatur).
Mollusken sind eine Gruppe von Tieren, die das Interesse einer breiten Vielfalt wissenschaftlicher Disziplinen halten. Trotz ihrer morphologischen Vielfalt 1, Artenreichtum ( an zweiter Stelle nach den Arthropoden in Bezug auf die Artenzahl 2) und der Relevanz für eine breite Palette von kommerziellen 3, medizinischen 4 und wissenschaftlichen Fragen 5-8, gibt es relativ wenige Mollusken – Arten , die zu behaupten kann , sein beide gut ausgestattete wissenschaftliche Modelle und einfach in einer Laborumgebung zu halten. Ein Mollusken , die viel von Disziplinen wie Neurobiologie 9 verwendet wird, Ökotoxikologie 10 und in jüngerer Zeit die Evolutionsbiologie 11,12, ist Spitzschlammschnecke, in erster Linie wegen seiner weiten Verbreitung und extrem einfache Wartung. Trotz seiner Popularität als "Modell" Organismus und seine lange Geschichte der Verwendung von Entwicklungsbiologen 13-19, die Reichweite und Leistung von molekularen Werkzeugen zur Verfügung, die L. StAGNalis wissenschaftliche Gemeinschaft liegt weit hinter der traditionellen Tiermodellen (Drosophila, Maus, Seeigel, Nematoden).
Unser Wunsch zu entwickeln Lymnaea als ein molekulares Modell von einem Interesse an den molekularen Mechanismen ergibt sich die Schalenbildung führen. Dies motiviert uns eine Reihe von Techniken zu verfeinern , die für die effiziente, konsistente und sensible Darstellung der Genexpression während der Lymnaea 's Entwicklung ermöglichen würde. Whole mount in situ Hybridisierung (WMISH) ist weit verbreitet für eine Vielzahl von Modellorganismen eingesetzt und ist seit mehr als 40 Jahren im Einsatz 20. In seinen verschiedenen Ausprägungen können, ISH räumlich eingesetzt werden, um spezifische Loci auf den Chromosomen, rRNA, mRNA und Mikro-RNAs zu lokalisieren.
Eine der Herausforderungen , mussten wir vor der Raffination eine WMISH Verfahren zur L. zu adressieren stagnalis war die Frage der sanft und effizient Embryonen und Larven von verschiedenen Stadien von t Extrahierener Ei-Kapseln, in denen sie abgelegt werden. Diese Extraktion oder "decapsulation 'muss effizient angemessene Material zu sammeln eine bestimmte in situ – Experiment, während zur gleichen Zeit die Aufrechterhaltung morphologischen und Zellintegrität um erreicht werden. Während andere Modellorganismen auch gekapselter Entwicklung durchlaufen, konnte erfolgreich in L. eingesetzt werden in unseren Händen keine der Methoden für diese Spezies berichtet stagnalis.
Die allgemeinen Ziele dieses Verfahrens sind daher: L. zu extrahieren stagnalis Embryonen und Larven aus ihren Kapseln in einem Hochdurchsatzverfahren , Pre-Hybridisierung Behandlungen anwenden, das den WMISH Signal zu optimieren, Embryonen und Larven mit zufriedenstellender WMISHsignals für die Bildgebung vorzubereiten.
Das hier beschriebene Verfahren ermöglicht die effiziente Visualisierung von RNA – Transkripte mit vermutlich unterschiedlichen Expressionsniveaus in allen Entwicklungsstadien der Spitzschlammschnecke.
