The goal of this protocol is to provide users with a set of methods for the high-throughput decapsulation of Lymnaea stagnalis embryos and larvae in preparation for whole mount in situ hybridization, and for subsequent pre- and post-hybridization treatments.
Montagem inteira de hibridização in situ (WMISH) é uma técnica que permite a resolução espacial das moléculas de ácidos nucleicos (ARNs mensageiros) muitas vezes dentro de uma preparação de tecido 'todo de montagem ", ou fase de desenvolvimento (tais como um embrião ou larva) de interesse. Os moluscos são um grupo de animais que mantêm o interesse de uma ampla diversidade de disciplinas científicas. Apesar da sua diversidade morfológica 1, riqueza de espécies (perdendo apenas para os artrópodes em termos de número de espécies 2) e relevância para uma ampla gama de comerciais 3, médicos e científicos 4 questões 5-8, há relativamente poucas espécies de moluscos que podem reivindicar a ser ambos os modelos científicos bem equipados e fácil de manter em um ambiente de laboratório. Esta extracção, ou 'desencapsulamento', tem de ser conseguida de forma eficiente, a fim de recolher o material adequado para uma determinada experiência in situ, enquanto, ao mesmo tempo mantendo a integridade morfológica e celular. Enquanto outros organismos modelo também submetidos a um desenvolvimento encapsulado, em nossas mãos nenhum dos métodos descritos para as espécies poderia ser empregada com sucesso em L. stagnalis.
Os objetivos gerais deste método são, portanto: para extrair L. stagnalis embriões e larvas de suas cápsulas em uma forma de alto rendimento, para aplicar tratamentos pré-hibridação que optimizam o sinal WMISH, para preparar embriões e larvas com WMISHsignals satisfatórios para a imagem latente.
O método descrito aqui permite a visualização eficiente de transcrições de RNA com presumivelmente diferentes níveis de expressão dentro de todos os estágios de desenvolvimento de Lymnaea stagnalis.
