Celler som vokser i en tredimensjonal (3-D) miljø representerer en markert forbedring i forhold til celledyrking i 2-D-miljøer (f.eks flasker eller retter). Her beskriver vi utviklingen av en flercellet 3-D organotypiske modell av det menneskelige tarmslimhinnen dyrket under mikrogravitasjons tilgjengelig ved å dreie-vegg-fartøy (RWV) bioreaktorer.
Fordi celler som vokser i en tredimensjonal (3-D) miljø har potensial til å bygge bro mange hull for celledyrking i 2-D-miljøer (f.eks., Kolber eller retter). Faktisk er det allment anerkjent at celler dyrket i kolber eller retter en tendens til å de-differensiere og mister spesialiserte funksjoner av vev fra der de ble hentet. For tiden er det i hovedsak to typer 3D-kultur systemer hvor cellene seeded inn stillaser etterligne de innfødte ekstracellulære matriks (ECM): (a) statiske modeller og (b) modeller som bruker bioreaktorer. Den første gjennombruddet var de statiske 3-D modeller. 3-D modeller som bruker bioreaktorer som roterende-vegg-fartøy (RWV) bioreaktorer er en nyere utvikling. Den opprinnelige konseptet av RWV bioreaktorer ble utviklet ved NASAs Johnson Space Center i begynnelsen av 1990 og antas å overvinne begrensninger av statiske modeller som for eksempel utvikling av hypoksiske, nekrotiske kjerner. De RWV bioreaktorer kan omgå ther problem ved å tilveiebringe fluiddynamikk som tillater effektiv diffusjon av næringsstoffer og oksygen. Disse bioreaktorer består av en rotator base som tjener til å støtte og rotere to forskjellige formater av kultur fartøy som avviker fra deres lufting kildetype: (1) Slow Turning Lateral Vessels (STLVs) med en koaksial oksygenator i sentrum, eller (2 ) Høy sideforhold Vessels (Harvs) med oksygenering via en flat, silikongummi gass overføring membran. Disse fartøyene tillate effektiv gassoverføring samtidig unngå bobledannelse og påfølgende turbulens. Disse betingelsene resulterer i laminær strømning og minimale skjærkraft som modellerer redusert gravitasjon (mikrogravitasjon) på innsiden av kulturen fartøyet. Her beskriver vi utviklingen av en flercellet 3-D organotypiske modell av tarmslimhinnen hos mennesker bestående av en intestinal epitelcellelinje og primære humane lymfocytter, endotelceller og fibroblaster dyrket under mikrogravitasjon levert av RWV bioreaktoren. </ P>
Den første gjennombrudd i byggingen av en 3-D-modell ble rapportert i begynnelsen av 1980-tallet da forskere begynt å undersøke forskjellige typer av stillaset (f.eks., Laminin, kollagen type I, kollagen IV, og fibronektin) og cocktail av vekstfaktorer for å forbedre celle-til-celle og ECM interaksjoner av "statisk" 3-D modeller 1-7. Siden den gang har det største problemet med disse modellene vært begrensninger i overføring av næringsstoffer og oksygen i løpet av de mellomstore og vev konstruerer 8. I motsetning til celler in in vivo miljø som mottar en jevn strøm av næringsstoffer og oksygen fra omgivende nettverk av blodårer, den statiske natur av disse modellene hindrer effektiv fordeling av dem til cellene. For eksempel vil celleaggregater som genereres i in vitro-modeller statiske som overstiger noen få millimeter i størrelse alltid utvikle hypoksiske, nekrotiske kjerner 9. De RWV bioreaktorer kan omgå dette problemetved å tilveiebringe fluiddynamikk som tillater effektiv diffusjon av næringsstoffer og oksygen 10-12. Men til dags dato, arbeid med RWV bioreaktorer har vært begrenset til inkludering av en eller to celletyper 13-17. Dessuten, i stedet for en romlig orientering som ligner naturlige vev, de celler som dannes celleaggregater. Hovedårsaken til disse begrensningene har vært mangelen på et stillas i stand til å innlemme celler i en integrert måte. Stillasene som brukes i RWV bioreaktorer til dags dato består, med få unntak 16-18, hovedsakelig av syntetisk mikroperler, rørformede sylindre eller små ark 13-15,19-23. Disse er stive materialer hvis sammensetning og fleksibilitet kan ikke manipuleres, og til hvilke celler som er festet til sin overflate. Således er det usannsynlig at disse modellene vil tilveiebringe et system for å evaluere, på en integrert måte, de forskjellige cellekomponenter slik som stromale celler (f.eks. Faktisk er det allment akseptert at ikke bare celle-til-celle, men også celle-til-ECM interaksjoner er kritisk for vedlikehold og differensiering av epitelceller i 3-D kultur modeller. Tidligere studier har vist at tarmen ECM-proteiner som kollagen I 24,36,37, laminin og fibronektin 38 39 er medvirkende til å påvirke intestinale epitelceller for å erverve romlig orientering som ligner den native mucosa. Således har utviklingen av nye teknologier, som vår 3-D-modell 24, som kan etterligne den fenotypiske mangfoldet av tarmen er nødvendig hvis forskere har til hensikt å gjenskape den komplekse cellulære og strukturell arkitekturog funksjon i tarmen mikromiljøet. Disse modellene representerer et viktig verktøy i utvikling og evaluering av nye orale legemidler og vaksinekandidater.
