The protocol described here details the induction of a hemogenic program in mouse embryonic fibroblasts via overexpression of a minimal set of transcription factors. This technology may be translated to the human system to provide platforms for future study of hematopoiesis, hematologic disease, and hematopoietic stem cell transplant.
This protocol details the induction of a hemogenic program in mouse embryonic fibroblasts (MEFs) via overexpression of transcription factors (TFs). We first designed a reporter screen using MEFs from human CD34-tTA/TetO-H2BGFP (34/H2BGFP) double transgenic mice. CD34+ cells from these mice label H2B histones with GFP, and cease labeling upon addition of doxycycline (DOX). MEFS were transduced with candidate TFs and then observed for the emergence of GFP+ cells that would indicate the acquisition of a hematopoietic or endothelial cell fate. Starting with 18 candidate TFs, and through a process of combinatorial elimination, we obtained a minimal set of factors that would induce the highest percentage of GFP+ cells. We found that Gata2, Gfi1b, and cFos were necessary and the addition of Etv6 provided the optimal induction. A series of gene expression analyses done at different time points during the reprogramming process revealed that these cells appeared to go through a precursor cell that underwent an endothelial to hematopoietic transition (EHT). As such, this reprogramming process mimics developmental hematopoiesis “in a dish,” allowing study of hematopoiesis in vitro and a platform to identify the mechanisms that underlie this specification. This methodology also provides a framework for translation of this work to the human system in the hopes of generating an alternative source of patient-specific hematopoietic stem cells (HSCs) for a number of applications in the treatment and study of hematologic diseases.
Hematopoies är en komplex utvecklingsprocess där hematopoetiska stamceller (HSCs) knoppas av hemogenic endotel närvarande i en mängd av embryonala hematopoietiska platser såsom Aorta-gonad-Mesonephros och placenta 1,2. Oförmågan att kulturen HSCs in vitro hindrar fördjupad analys av denna process samt den kliniska tillämpningen av dessa studier. Den nya arbete som fastställts av Yamanaka och kollegor för att generera inducerade pluripotenta stamceller (iPSCs) från somatiska fibroblaster provides en ram för transkriptionsfaktor (TF) baserade överuttryck strategier omprogrammering cell öde 6,7. Detta arbete har lett forskare inom flera områden för att generera celltyper av val via TF omprogrammering av lätt erhållas somatiska celler. Det första steget för att säkerställa korrekt omprogrammering i denna mus systemet var att utveckla en reporter linje som fungerade som en utläsning för CD34 uttryck, en känd markör i endotelceller progenitorceller och HSCs. För att göra detta har de huCD34-TTA och TetO-H2BGFP transgen mus linjer som används för att generate dubbel transgen mus embryonala fibroblaster (MEF), nu betecknat 34 / H2BGFP, som fluorescerar grönt vid aktivering av CD34-promotom 8. Detta tillät screening av en mängd olika TF är kända för att krävas vid olika tidpunkter under hematopoetisk specifikation och utveckling. Från och med 18 TF i PMX retrovial vektorer (bestämd genom litteratur utvinning och profilering av GFP etikettkvarhållande HSCs från den tidigare beskrivna 34 / H2BGFP möss), 34 / H2BGFP MEF transducerades med alla faktorer och odlades på AFT024 HSC bärande stromaceller. Efter upptäckt av 34 / H2BGFP aktivering var TF därefter avlägsnas från omprogrammering cocktail tills den optimala uppsättningen av TF för reporter aktivering identifierades. Efter denna inledande skärm, var de faktorer som överförts till en DOX inducerbar pFUW vektorsystem för att möjliggöra reglerbar uttryck för TF. Eftersom dessa två DOX reglerbara system är oförenliga (34 / H2BGFP celler och pFUW inducerbara vektorer), MEF frånvildtyp C57BL / 6-möss som krävs. Det var också nödvändigt att skapa en lämplig mikromiljö för att tillåta hemogenesis att gå vidare och skapa multilineage klonogena stamceller.
Aktuella studier försöker programmera somatiska celler i hematopoetiska stamceller och progenitorceller (HSPCs) har träffat olika nivåer av framgång 9-11. Hittills har alstringen av både mus- och humana transplanterbara HSPCs med lång sikt och självförnyande repopulerande förmåga inte uppnåtts med användning av samma uppsättning av TF: er. I detta protokoll, ger vi en detaljerad beskrivning av den tidigare fastlagda strategin att reproducerbart framkalla hemogenesis i MEF. Vi visar att införandet av en minimal uppsättning TF (Gata2, Gfi1b, cFos och Etv6) är i stånd att anstiftan ett komplext utvecklingsprogram in vitro som ger en plattform genom vilken utvecklings hematopoes och klinisk tillämpning av hematopoetisk omprogrammering kan studeras närmare 12.
