Nous présentons ici un protocole pour mesurer la force des interactions entre une surface inorganique bien définie et soit des peptides ou des acides aminés par une seule molécule des mesures de spectroscopie de force au moyen d'un microscope à force atomique (AFM). Les informations obtenues à partir de la mesure est importante pour mieux comprendre le matériau interphase peptide-inorganique.
Les interactions entre les protéines ou peptides et des matériaux inorganiques conduisent à plusieurs procédés intéressants. Par exemple, en combinant des protéines avec des minéraux conduit à la formation de matériaux composites ayant des propriétés uniques. En outre, le procédé de l'encrassement biologique indésirable est initiée par l'adsorption des molécules biologiques, principalement des protéines, sur les surfaces. Cette couche organique est une couche d'adhérence pour les bactéries et leur permet d'interagir avec la surface. Comprendre les forces fondamentales qui régissent les interactions à l'interface organique-inorganique est donc important pour de nombreux domaines de la recherche et pourrait conduire à la conception de nouveaux matériaux pour des applications optiques, mécaniques et biomédicales. Ce document démontre une technique de spectroscopie de force une seule molécule qui utilise un AFM pour mesurer la force d'adhérence entre soit des peptides ou des acides aminés et des surfaces inorganiques bien définies. Cette technique implique un protocole de fixation de la molécule biologique à l'AFMpointe à travers une liaison flexible covalente et une seule molécule des mesures de spectroscopie de force par microscope à force atomique. En outre, une analyse de ces mesures est inclus.
L'interaction entre les protéines et les minéraux inorganiques conduit à la construction de matériaux composites ayant des propriétés particulières. Cela comprend des matériaux à haute résistance mécanique ou des propriétés optiques uniques. 1, 2, par exemple, la combinaison de la protéine de collagène avec l'hydroxyapatite minérale génère soit des os mous ou durs pour les différentes fonctionnalités. 3 peptides courts peuvent également lier des matériaux inorganiques avec une grande spécificité. 4, 5, 6 La spécificité de ces peptides a été utilisé pour la conception de nouveaux matériaux magnétiques et électroniques, 7, 8, 9 fabrication des matériaux nanostructurés, croissance de cristaux, 10 et la synthèse de nanoparticules. 11 Comprendre le mécanisme sous – jacent des interactions entre des peptides ou des protéines et des matières inorganiques va donc nous permettre de concevoir de nouveaux matériaux composites avec l' amélioration des propriétés d' adsorption. En outre, depuis l'interphase des implants avec une réponse immunitaire est médiée par des protéines, une meilleure compréhension des interactions des protéines avec des matériaux inorganiques permettra d'améliorer notre capacité à concevoir des implants. Un autre domaine important qui implique des protéines interagissant avec des surfaces inorganiques est la fabrication de matériaux anti-salissures. 12, 13, 14, 15 encrassement biologique indésirable est un processus dans lequel les organismes se fixent sur une surface. Il a de nombreuses conséquences néfastes sur nos vies. Par exemple, l'encrassement biologique des bactéries sur les dispositifs médicaux conduit à des infections nosocomiales. Biofouling des organismes marins sur les bateaux et les grands navires augmente la consommation de carburant. 12, 16, 17, 18
Simple-molécule vigueur spectroscopie (SMFS), en utilisant un AFM, peut directement mesurer les interactions entre un acide aminé ou un peptide avec un substrat. 19, 20, 21, 22, 23, 24, 25, 26 D' autres méthodes telles que l' exposition sur phage, 27, 28 microbalance à cristal de quartz (QCM) 29 ou par résonance plasmonique de surface (SPR) 29, 30, 31, 32,ref "> 33 mesure les interactions des peptides et des protéines à des surfaces minérales en vrac. 34, 35, 36 Cela signifie que les résultats obtenus par ces procédés se rapportent à des ensembles de molécules ou d'agrégats. Dans SMFS, un ou très peu de molécules sont fixées à la pointe de l'AFM et de leurs interactions avec le substrat désiré est mesurée. Cette approche peut être étendue à l'étude du repliement des protéines en tirant sur la protéine à partir de la surface.
