Denne artikel indeholder en forenklet og standardiseret protokol til induktion af depressiv-lignende adfærd i kronisk immobiliseret mus ved hjælp af en fastholdelses. Desuden, adfærd og fysiologiske teknikker til at kontrollere induktion af depression forklares.
Depression er endnu ikke fuldt forstået, men forskellige årsagsfaktorer er blevet rapporteret. For nylig, forekomsten af depression er steget. Men, terapeutiske behandlinger for depression eller forskning på depression er knappe. Således i det foreliggende papir, foreslår vi en musemodel af depression induceret af bevægelse restriktion. Kronisk mild stress (CMS) er en velkendt teknik til at fremkalde depressiv-lignende adfærd. Det nødvendiggør imidlertid en kompleks procedure, der består af en kombination af forskellige milde belastninger. I modsætning hertil er kronisk immobiliserings stress (CIS) en let tilgængelig kronisk stress model, modificeret fra en fastholdelses model, der inducerer depressiv adfærd ved at begrænse bevægelse ved hjælp af en fastholdelses i en vis periode. For at evaluere depressiv-lignende adfærd, den saccharose præference test (SPT), hale suspension test (TST), og ELISA assay til at måle stress markør kortikosterone niveauer kombineres i det nuværende eksperiment. De beskrevne protokoller illustrerer induktion af CIS og evaluering af ændringer i adfærd og fysiologiske faktorer for validering af depression.
Major depressive Disorder (MDD) er den førende årsag til psykisk handicap på verdensplan, med en incidens, der er stigende hurtigere end forventet. I 2001, Verdenssundhedsorganisationen forudsagde, at MDD ville være den næstmest almindeligt forekommende sygdom i verden af 2020. Men, det var allerede den næstmest almindeligt i 20131. Desuden, nuværende antidepressiva har mange begrænsninger, herunder forsinket effektivitet, Drug resistens, tilbagefald, og forskellige bivirkninger2,3. Forskerne skal derfor udvikle mere effektive antidepressiva. Men den tvetydige Patofysiologi af MDD udgør en hindring for udviklingen af nye antidepressiva.
Langsigtet stress er den vigtigste risikofaktor for MDD. Det kan fremkalde dysfunktion i hypothalamus-hypofyse-adrenal (hPa) akse, som også er relateret til MDD ætiologi4,5. Som tidligere beskrevet spiller hPa-aksen en afgørende rolle i stress-induceret psykiatrisk Patofysiologi, herunder depression og angstlidelser ved at øge kortikosterone niveauer6,7,8, 9. mange dyremodeller har været baseret på vedvarende aktivering af hPa-aksen, som observeres hos patienter med MDD4. Desuden, høje glukokortikoider induceret af kronisk stress og subkutant injiceres glukokortikoider forårsage depressiv adfærd sammen med neurale celledød, atrofi af neuronal processer, og reduceret voksne neurogenese i hjernen af gnavere10 , 11. en anden vigtig hjerne område forbundet med depression er den mediale præfrontale cortex (mpfc). Den mpfc spiller en afgørende rolle i at kontrollere hjernens subregioner, såsom hypothalamus og amygdala, at kontrollere følelsesmæssige adfærd og stress svar8,9. For eksempel, læsioner i dorsale mpfc induceret hPa akse dysfunktion og øget kortikosteron sekretion på grund af tilbageholdenhed stress12,13. En nylig undersøgelse viste også, at gentagen tilbageholdenhed stress øget kortikosterone niveauer, som kunne reduceres ved glutamin tilskud via glutamat-glutamin cyklus mellem neuroner og astrocyt i mPFC9.
Den første kroniske stress paradigme bruges til at studere ætiologi af MDD blev foreslået af Katz14. Willner et al. derefter foreslået en kronisk mild stress (CMS) model baseret på resultaterne af Katz. De bekræftede, at modellen havde prædiktiv gyldighed ved at observere, at antidepressiva restaureret CMS-induceret anhedonic-lignende adfærd15,16. Typisk består CMS-modellen af en kombination af forskellige milde belastninger, såsom mild støj, bur vippe, vådt sengetøj, ændrede lys-mørke cyklusser, bur ryster, tvungen svømning, og sociale nederlag. CMS-modellen udnyttes i vid udstrækning af forskere; men denne model er af dårlig replikbarhed, og tid-og energi-ineffektive. Derfor er der en stigende efterspørgsel efter en standardiseret og forenklet protokol for induktion af depressiv-lignende adfærd og fysiologiske analyse til at evaluere depression. Sammenlignet med CMS-modellen, den kroniske immobilisering stress (CIS; også kendt som kronisk tilbageholdenhed stress) model er enklere og mere effektiv; Derfor kan CIS-modellen anvendes i vid udstrækning i kroniske stressundersøgelser17,18,19,20,21,22,23, 24. Desuden kan CIS anvendes i både mandlige og hunmus til at udvikle depressiv adfærd25,26. Under CIS, dyr er placeret i en krop-fit størrelse cylinder til 1-8 timer om dagen for 2 eller 4 uger9,27,28. Af disse, tilbageholdenhed stress betingelse for 2 timer om dagen for 2 uger er tilstrækkelig til at forårsage depressiv adfærd med minimal smerte i mus9,28. Under fastholdelses betingelser blev blod kortikosterone-niveauerne hurtigt forhøjet9,28,29. Flere undersøgelser har vist, at CIS-modellen har forudsigende gyldighed, hvilket bekræfter, at CIS-induceret depressive-lignende symptomer er genoprettet ved antidepressiva19,20,30,31. Kompleksiteten af hjernen og heterogeniteten af MDD gør det udfordrende at skabe dyremodeller, der fuldstændig gengiver betingelsen.
