开源高内涵分析利用自动荧光寿命成像显微镜

我们目前使用的测定使用荧光共振能量转移（FRET）的读数蛋白质相互作用自动荧光寿命成像（FLIM）一个开源的高内涵分析（HCA）的仪器。此openFLIM-HCA仪数据采集是通过软件写入μManager和数据分析在FLIMfit进行控制。
该程序的总体目标是演示如何使用开源软件和基本仪器在自动化多孔板工作流程中实施时间门控荧光寿命成像显微镜 （FLIM）。该方法可应用于一系列固定或活细胞中基于 FLIM 的读数。它对于读取细胞信号处理或蛋白质相互作用特别有用。

然后检查显示的输出跟踪。并设置航向延迟时间，使完整的仪器响应功能配置文件处于快速延迟盒的扫描范围内。调整 NEUTRAL DENSITY 和 EXPOSURE TIME 参数。
以及帧的累积，使相机在亮度时间门中不饱和且有效积分时间不小于 200 毫秒。在 FLIM 控制菜单下，选择 Fast Delay 框并在整个延迟范围内设置 25 皮秒延迟步长。接下来，选择 populate delays（填充延迟）并单击 snap FLIM image（捕捉 FLIM 图像）以获取仪器响应函数图像。
并等待收购完成。在光路控制菜单下，选择 neutral density（中性密度），然后单击 blocked（阻挡）以阻挡激光。打开 FLIM 控制菜单并选择 fast delay 框，通过增加增量框中的值来减少延迟步数。
然后单击 snap FLIM image 以获取背景相机图像。要获取参考晶粒数据，请转到 XYZ 控制选项卡，然后在 go to well 对话框中输入 H4 井。接下来，转到光路控制选项卡，选择合适的激发、二向色和发射滤光片，以便在快速延迟框的整个范围内使用查找最大点功能对 M 绿松石荧光蛋白进行成像。

然后点击 AF now 启动自动对焦程序。偏移值将出现在焦点偏移自动对焦字段上。在 AF 偏移窗口中输入此偏移值，然后单击 AF now 两次以重复自动对焦过程。
偏移值现在应设置为零，表示自动对焦系统正常运行。接下来，在 FLIM control 选项卡下，选择自动门控功能以确保延迟点的对数采样。将 accumulate frames 设置为可产生所需总数据采集时间的值。

总体而言，每个孔中所有视场的平均寿命在多孔板中有所不同。对 A1 孔中成像的所有细胞的供体荧光强度衰减曲线进行平均，并与单指数衰减模型和仪器响应函数拟合，表明拟合模型是有效的。通过像素级单指数拟合获得的多孔板每列的平均荧光寿命的进一步分析表明，接头长度较短的 Fret 构建体的寿命缩短，模拟了具有不同 Fry 效率的实验。
在本实验中，不同抑制剂对蛋白质寡聚化作用的 fret 测定的 FLIM 数据与单个指数衰减模型拟合。荧光寿命板图说明了 8 个视场的平均每孔平均寿命。一个小集合，每孔四个视野成像，而我们的 FLIM 分析可以在大约两个半小时内完成，包括获取仪器响应功能、参考芯片数据和以 FLIM 拟合运行分析的时间。
