Makromolekylära handel mellan växtceller kan bedömas genom att tillfälligt uttrycka en fluorescerande-taggade proteinet av intresse samt en analys av intra-och intercellulära distribution av konfokalmikroskopi.
Abstract
Här presenterar vi ett enkelt och snabbt protokoll för att upptäcka och bedöma omfattningen av cell-till-cell makromolekylära transporter i Planta. I detta protokoll är en fluorescerande taggad-proteinet av intresse kortvarigt uttryck i växtvävnad efter biolistic leverans av sin kodning DNA-konstruktion. Intra-och intercellulära distribution av de märkta proteinet analyseras sedan av konfokalmikroskopi. Vi beskriver denna teknik i detalj, vilket ger steg-för-steg-protokoll för att analysen och bedöma omfattningen av symplastic protein transport i tre växtarter, Arabidopsis thaliana, Nicotiana benthamiana och N. tabacum (tobak).

Bakgrund Symplastic transport av makromolekyler genom anläggningen intercellulära anslutningar, plasmodesmata, är av intresse för många växt-patologer och biologer. Discussion
Nyckeln till framgång för symplastic transporter analysen är att med hög omvandlingseffektiviteten, som möjliggör produktion av statistiskt signifikanta och lätt spårbar signal kluster. Detta kan uppnås genom att använda bladen skördas från friska, robusta växter och förbereda guldpartiklar belagda med en ren och koncentrerad DNA-förberedelser.
