Phase-shift nanoemulsions (PSNE) kan fordampes ved hjælp af høj intensitet fokuseret ultralyd til at forbedre lokal opvarmning og forbedre termisk ablation i tumorer. I denne rapport, er fremstillingen af stabile PSNE med en snæver størrelsesfordeling beskrevet. Endvidere er effekten af fordampet PSNE på ultralyd-medieret ablation påvist i væv der efterligner fantomer.
Høj intensitet fokuseret ultralyd (HIFU) anvendes klinisk til termisk ablatere tumorer. At øge lokal opvarmning og forbedre termisk ablation i tumorer, er lipid-overtrukne perfluorcarbongasser smådråber blevet udviklet, som kan fordampes ved HIFU. Vaskulaturen i mange tumorer er unormalt utæt på grund af deres hurtige vækst og nanopartikler kan trænge ind i fenestrationer og passivt akkumuleres i tumorer. Således kan styring af størrelsen af smådråberne resultere i bedre akkumulering i tumorer. I denne rapport er fremstillingen af stabile dråber i en faseforskydning nanoemulsion (PSNE) med en snæver størrelsesfordeling beskrevet. PSNE blev syntetiseret ved sonikering af en lipidopløsning i nærværelse af flydende carbonperfluorid. En snæver størrelsesfordeling blev fremstillet ved at ekstrudere de PSNE flere gange under anvendelse af filtre med porestørrelser på 100 eller 200 nm. Størrelsesfordelingen blev målt over en periode på 7 dage ved hjælp af dynamisk lysspredning. Polyacrylamide hydrogeler indeholdende PSNE blev fremstillet til in vitro forsøg. PSNE smådråber i hydrogelerne blev fordampet med ultralyd og de resulterende bobler forbedret lokal opvarmning. Fordampet PSNE muliggør hurtigere opvarmning og reducerer også ultralyd intensitet nødvendig for termisk ablation. Således er PSNE forventes at forbedre termisk ablation i tumorer, potentielt forbedre terapeutiske resultater af HIFU-medierede termisk ablation behandlinger.
