Plant virale nanodeeltjes (VNPs) zijn veelbelovend platform voor toepassingen in de medische biologie. Hier beschrijven we de procedures voor plantaardige VNP vermeerdering, zuivering, karakterisering, en bioconjugatie. Ten slotte tonen we de toepassing van VNPs voor tumor homing en beeldvorming met behulp van een muis xenograft model en fluorescentie beeldvorming.
Het gebruik van nanomaterialen heeft de potentie om materialen wetenschap en geneeskunde een revolutie. Momenteel worden verschillende nanodeeltjes onderzocht voor toepassing in beeldvorming en therapie. Virale nanodeeltjes (VNPs) afgeleid van planten kan worden beschouwd als zelf-geassembleerde bionanomaterials met verschillende grootten en vormen. Plantenvirussen onderzochte in de Steinmetz lab zijn icosahedrale deeltjes gevormd door Cowpea mosaic virus (CPMV) en Brome mosaic virus (BMV), die beide 30 nm in diameter. We ontwikkelen ook staafvormig en filamenteuze structuren afgeleid van de volgende plantenvirussen: Tobacco mosaic virus (TMV), die stijve staven vormen met afmetingen van 300 nm met 18 nm en aardappelvirus X (PVX) die draadvormige deeltjes vormen 515 nm lang en 13 nm breed (wordt verwezen naar refs. 1 en 2 voor meer informatie over VNPs).
<p class="jove_content"> Vanuit het oogpunt van een materiaal wetenschapper's standpunt, VNPs zijn aantrekkelijk bouwstenen om verschillende redenen: de deeltjes zijn monodisperse, kan worden geproduceerd met gemak op grote schaal in planta, zijn uitzonderlijk stabiel en biocompatibel zijn. Ook VNPs zijn "geprogrammeerd" eenheden, die specifiek kunnen worden gemanipuleerd met behulp van genetische modificatie of chemische methoden bioconjugatie 3. De structuur van VNPs is bekend dat atomaire resolutie, en wijzigingen kunnen worden uitgevoerd met ruimtelijke precisie op het atomaire niveau 4, een niveau van controle die niet kan worden bereikt met behulp van synthetische nanomaterialen met de huidige state-of-the-art technologieën.In dit artikel beschrijven we de verspreiding van CPMV, PVX, TMV, en BMV in Vigna ungiuculata en Nicotiana benthamiana planten. Extractie en zuivering protocollen voor elke VNP gegeven. Methoden voor het karakteriseren van gezuiverd en chemisch gelabelde VNPs beschreven. In dit onderzoek richten we ons op chemical etikettering van VNPs met fluoroforen (bijv. Alexa Fluor 647) en polyethyleenglycol (PEG). De kleurstoffen vergemakkelijken volg-en detectiesysteem van de VNPs 5-10, en PEG vermindert immunogeniciteit van het eiwitachtige nanodeeltjes terwijl het verbeteren van hun farmacokinetiek 8,11. We laten zien tumor homing van gePEGyleerde VNPs gebruik van een muis xenograft tumor model. Een combinatie van fluorescentie beeldvorming van weefsels ex vivo met Maestro Imaging System, fluorescentie kwantificering in gehomogeniseerde weefsels, en confocale microscopie wordt gebruikt om biodistributie te bestuderen. VNPs worden gewist via het reticulo-endotheliale systeem (RES); tumor homing wordt passief bereikt via de verbeterde doorlaatbaarheid en retentie (EPR) effect 12. De VNP nanotechnologie is een krachtige plug-and-play-technologie om beeld en te behandelen plaatsen van de ziekte in vivo. Wij blijven VNPs van drugs ladingen en klinisch relevante imaging eenheden, alsmede weefsel-specifieke liganden dragen naargericht moleculaire receptoren tot overexpressie gebracht in kanker en cardiovasculaire aandoeningen.
Dit protocol verschaft een benadering voor de chemische modificatie van VNPs en hun toepassingen in vivo tumor imaging. Het dier fluorescentie beeldvorming, fluorescentie kwantificering en immunohistochemie technieken hier gepresenteerd zijn nuttig voor het bestuderen van biologische verdeling en evalueren van tumor homing.
