We genetisch coderen natuurlijk aminozuur, p-azido-L-fenylalanine op verschillende gerichte posities in GPCRs en tonen de veelzijdigheid van de azidogroep in verschillende toepassingen. Deze omvatten een gerichte fotoverknoping technologie om resten in de ligand-bindingsplaats van een GPCR te identificeren, en de site-specifieke bioorthogonal modificatie van GPCR's met een peptide-epitoopmerker of fluorescente probe.
Structurele en dynamische studies van G-eiwit gekoppelde receptor (GPCR) signalering complexen te vergemakkelijken, zijn nieuwe benaderingen nodig zijn om informatieve probes of labels introduceren in uitgedrukt receptoren die geen receptor functie niet verstoren. We gebruikten amber codon suppressie technologie genetisch coderen van het niet-natuurlijke aminozuur, p-azido-L-fenylalanine (azf) bij verschillende gerichte posities in GPCRs heteroloog tot expressie gebracht in zoogdiercellen. De veelzijdigheid van de azidogroep is hier weergegeven in verschillende toepassingen GPCR bestuderen in hun natieve cellulaire omgeving of in detergens gesolubiliseerde omstandigheden. Ten eerste tonen we een cel-gebaseerde gerichte fotoverknoping technologie aan de residuen in de ligand-bindingsplaats van GPCR wanneer een tritium-gelabeld kleine moleculen ligand verknoopt een genetisch gecodeerd aminozuur azido identificeren. We tonen dan plaatsspecifieke modificatie van GPCR's door de bioorthogonal Staudinger-Bertozzi ligatie reactiestie dat de azidogroep richt met behulp van fosfine derivaten. We bespreken een algemene strategie voor gerichte peptide-epitoop tagging van uitgedrukt membraaneiwitten in-cultuur en de detectie met behulp van een whole-cell-gebaseerde ELISA aanpak. Tot slot laten we AZF-GPCR selectief kan worden gelabeld met fluorescente probes. De methoden besproken algemeen, aangezien zij kunnen in beginsel worden toegepast op elk aminozuur positie in een GPCR tot expressie actieve signalering complexen ondervragen.
Heptahelical G-proteïne gekoppelde receptoren (GPCR's) omvatten een superfamilie van zeer dynamische membraaneiwitten die belangrijk en diverse extracellulaire signalen mediëren. In het klassieke paradigma wordt receptoractivering gekoppeld ligand geïnduceerde conformatieveranderingen. 1, 2 hebben recente ontwikkelingen in de structurele biologie van GPCRs belangrijke inzicht in de moleculaire mechanismen van transmembraan signalering ontvangen. 3-5 echter te begrijpen met een grotere nauwkeurigheid de chemische werkingsmechanisme en structurele dynamica van GPCR-signalering, is een toolkit van benaderingen nodig zijn om informatieve moleculaire en chemische probes te nemen aan actieve signalering complexen ondervragen.
Daarom passen wij een methode om plaatsspecifiek voeren niet of minimaal storende sondes in expressie receptoren gebaseerd op natuurlijk aminozuur (UAA) mutagenese met amber codon suppressie technologie eerder ontwikkeld door Schultz en coworkers. 6 We geoptimaliseerd de UAA mutagenese methode om een high-yield expressie en mutagenesesysteem voor eiwitten zoals GPCR's, die moeilijk uit te drukken in een andere dan de zoogdiercellen meeste heterologe systemen zijn te bereiken. Met behulp van een orthogonale gemanipuleerde suppressor tRNA en evolueerde aminoacyl-tRNA synthetase paar voor een specifiek UAA, we plaatsspecifiek geïntroduceerd UAAs in uitgedrukt doel GPCR's. De succesvolle integratie van UAAs, p-acetyl-L-fenylalanine (ACF), p-benzoyl-L-fenylalanine (BZF), en p-azido-L-fenylalanine (azf) is aangetoond in ons model GPCRs – rhodopsine en menselijke CC chemokine receptor CCR5. 7, 8
In beginsel kan een UAA genetisch gecodeerd op elke positie in het eiwit sequentie en deze woning is een waardevol biochemische instrument omdat het zorgt voor single-codon scannen van een doel GPCR. We richten ons hier specifiek op de veelzijdigheid van de UAA, AZF, die een reactieve Azi heeftdo groep. Naast het dienen als een uniek infrarood (IR) sensor, 8, 9 azf kan ook dienen als een fotoactiveerbare verknoper door reactie met naburige primaire aminen of alifatische waterstofatomen. Bovendien kan het biologisch inerte azidogroep nemen als een selectief chemisch handvat bioorthogonal labelingsreacties. Hier laten we voorbeelden illustreren de nuttige toepassingen van plaats-specifieke incorporatie van AZF in GPCRs, zoals gerichte fotoverknoping te controleren een receptor-ligand-complex en modificatie van GPCR door bioorthogonal epitoop tagging en fluorescente labeling strategieën.
Fotoactiveerbare reagentia zijn gebruikt om biologische systemen te bestuderen sinds 1960. 10 In deze periode zijn een overvloed aan receptor-ligand verknoping experimenten gerapporteerd dat GPCR complexen studie, waarvan de meeste betrekking het gebruik van foto-affiniteit liganden. 11, 12 Echter, deze toepassingen zijn technisch beperkt, aangezien zij require synthese van liganden richting een verknopende groep. 13-15 bovendien de crosslinker deel in het ligand, is het een uitdaging om de locatie van de verknoping bepalen de GPCR. Plaatsspecifiek introduceren fotoverknopende groepen UAAs in eiwitten met het amber codon onderdrukking technologie is een waardevolle vooruitgang. 16 17 We ontwikkelden een fotoverknoping techniek om de binding interface op een receptor die betrokken is bij de vorming van een receptor-ligand-complex te identificeren levende cellen door de invoering fotolabiele groepen in GPCR's. 18, 19 Hier beschrijven we de experimentele protocol en de wijze van data-analyse voor de toepassing van deze gerichte fotoverknoping technologie om de bindingsplaats van een klein-molecule ligand, tritiumhoudend maraviroc, op CCR5 identificeren. Deze werkwijze in op de precieze kwantificering van de radioactieve handvat op het ligand, naast behoud van de natuurlijke chemische structuur van het ligand.
Fluorescentie-gebaseerde technieken ondersteunen juist begrip van de structurele basis van receptoractivering door direct sonderen van de conformationele toestand van de receptor. 20, 21 echter technieken bezit de flexibiliteit om fluorescerende labels introduceren in GPCRs plaatsspecifiek beperkt. Wij zijn geïnteresseerd in het gebruik van bioorthogonal chemische modificatie strategieën om single-molecule detectie (SMD) van GPCR signalering complexen te vergemakkelijken. 22 De azidogroep kan deelnemen aan bioorthogonal chemicaliën zoals Staudinger-Bertozzi ligatie, 23, 24 kopervrije-stam bevorderd azide- alkyne cycloadditie (SpAAC) 25 en-koper-gekatalyseerde azide-alkyn cycloadditie (CuAAC). 26 Wij richten ons hier op de Staudinger-Bertozzi ligatie reactie die de specifieke reactie tussen een azide en een fosfine gaat. We demonstreren het gebruik van twee verschillende fosfine derivaten geconjugeerd aan een peptide epitoop (FLAG peptide) of een fluorescerend label(Fluoresceïne) naar plaatsspecifieke modificatie van GPCR's te bereiken.
We hebben eerder geoptimaliseerd voorwaarden voor plaats-specifieke labeling van rhodopsine AZF varianten met de röntgenbron kristalstructuur en dynamische simulaties doelplaatsen die aan oplosmiddel blootgestelde zijn kiezen. 27 28 We geïllustreerd tevens de mogelijkheid om zonder achtergrond labeling bereiken door Staudinger- Bertozzi ligatie. 29 We tonen hier de algemene procedure gebruikt voor fluorescente labeling van een detergens gesolubiliseerde receptor die is geïmmobiliseerd op een matrix immuno vervolgens gevisualiseerd door in-gel fluorescentie bereiken. Daarnaast tonen we een nuttige uitbreiding op deze etikettering strategie om posities ontvankelijk voor etikettering op een receptor van onbekende structuur, CCR5 identificeren. Dit wordt uitgevoerd met een gerichte peptide-epitoop tagging strategie die is gebaseerd op bioorthogonal modificatie van AZF-GPCR varianten in een celgebaseerde semi-high throughput format. 30 Ditmethode maakt gebruik van de multi-step detectie eigenschappen van een cel-oppervlak ELISA etikettering gebeurtenissen volgen.
De methoden die we hier besproken zijn algemeen en kunnen in principe worden toegepast op elk GPCR opgenomen met AZF met het amber codon onderdrukking technologie. In de hier gepresenteerde gedetailleerde weergave van de stappen die in zoogdiercellen expressie van receptoren opgenomen met UAAs zoals AZF met het onnatuurlijke aminozuur mutagenese werkwijze en de latere toepassingen structurele en dynamische studies van GPCRs vergemakkelijken protocollen.
We beschrijven hier een robuuste methode voor plaatsspecifieke integratie van een reactieve probe, AZF, in GPCRs en tonen drie nuttige toepassingen van dit hulpmiddel om de structuur en dynamiek van GPCR bestuderen. Onze methode naar site-specifiek te nemen UAAs omzeilt een fundamenteel probleem met een alternatieve strategie op basis van chemisch labelen 32, 33 of vastmaken fotocrosslinkers 34 tot een enkel toegankelijk cysteïne mutant.
