Hepatitis C Virus (HCV) is a major human pathogen that causes liver disorders, including cirrhosis and cancer. An HCV infectious cell culture system is essential for understanding the molecular mechanism of HCV replication and developing new therapeutic approaches. Here we describe a protocol to investigate various stages of the HCV replication cycle.
Hepatitis C virus (HCV) påvirker 3% af verdens befolkning og forårsager alvorlige leversygdomme, herunder kronisk hepatitis, cirrhose og hepatocellulært carcinom. HCV er en kappebærende RNA-virus, der tilhører familien Flaviviridae. Nuværende behandling ikke er fuldt effektiv og forårsager bivirkninger. Der er ingen HCV-vaccine til rådighed. Således vedvarende indsats er påkrævet for at udvikle en vaccine og bedre behandling. En HCV cellekultur-system er afgørende for at studere forskellige stadier af HCV vækst, herunder viral indgang, genomreplikation, emballering og påstigning. I den nuværende fremgangsmåde fremlagt, vi anvendte en vildtype intragenotype 2a kimære virus, FNX-HCV og en rekombinant FNX-Rluc virus bærer et Renilla-luciferase-reportergen for at undersøge virusreplikation. En human hepatomacellelinie (Huh-7-baseret) blev anvendt til transfektion af in vitro-transkriberet HCV genomiske RNA'er. Cellefrie kultursupernatanter, proteinlysater og total RNA blev høstet på forskellige tidspunkter efter transfektion til at vurdere HCV-vækst. HCV-genomet replikation status blev vurderet ved kvantitativ RT-PCR og visualisere tilstedeværelsen af HCV dobbeltstrenget RNA. HCV protein udtryk blev bekræftet ved Western blot og immunofluorescensassays anvender antistoffer specifikke for HCV NS3 og NS5A proteiner. HCV-RNA-transficerede celler frigivet infektiøse partikler i dyrkningssupernatanten, og den virale titer blev målt. Luciferaseassays blev benyttet til at vurdere replikation niveau og infektivitet af reporter HCV. Afslutningsvis præsenterer vi forskellige virologiske analyser til karakterisering af forskellige stadier af HCV-replikation cyklus.
Hepatitis C virus (HCV) forårsager skrumpelever og leverkræft. Det påvirker 170 millioner mennesker på verdensplan med 350.000 mennesker dør hvert år 1-3. HCV er et positiv-strenget RNA-virus med et genom størrelse på 9,6 kb. HCV-genomet er oversat som en enkelt polyprotein på ~ 3.000 aminosyrerester, proteolytisk spaltes af forskellige cellulære og virale proteaser i 10 polypeptider. HCV er prototypen virus i slægten Hepacivirus og tilhører familien Flaviviridae 4. Ved eksponering HCV etablerer kronisk infektion i 80% af individerne. Infektionen er oftest asymptomatisk og rettidig diagnose kan tillade terapeutisk intervention for at forhindre leveren forringelse. Nuværende behandling er suboptimal og ingen vaccine er tilgængelig 5,6.
Ætiologien af hepatitis C blev først beskrevet i 1989 7.. Studere HCV-replikation er vigtigt for hepatitis C-vaccine og behandling forskning, men det havde væretlænge hæmmet af manglen på en effektiv viral kultur system. En molekylær klon af HCV blev vist at være smitsomme i chimpanser upon intrahepatisk podning 8. Efterfølgende blev HCV Subgenomiske replikoner beskrevet som tillod at dissekere det virale genom replikation fase i et cellekultursystem 9,10. Opdagelsen af en genotype 2a HCV isolere JFH-1 (japansk Fulminant hepatitis-1), er i stand til at inficere cellekultur åbnet nye muligheder for HCV-replikation forskning 11-13. Genotype 2a stamme JFH-1 baseret inter-og intra-genotypiske kimære virus og genotype 1 HCV baserede smitsomme kultur systemer er tilgængelige samt 14-18.
Vi har med succes brugt JFH-1 stamme og HCV intragenotype 2a kimære virus at opnå høj opløsning funktionelle profilering kort af protein domæner og cis-virkende RNA elementer 19,20. Ifølge dette, her beskriver vi en effektiv dyrkningssystem rutinemæssigt anvendes, der tilladerstudere forskellige stadier af HCV replikationscyklussen og vært-patogen interaktion.
